| BackgroudInvation and metastasis are the two biological characteristics of malignant tumors.Accordingly,it is one of the effective means to control the tumor,improve the cure rate and extend to the survival time of patients is to prevent the invasion and metastasis of malignant tumors.The occurrence of invation and metastasis is a serial process,including the degradation of basement membrane of tumor cells to invade surrounding tissues,tumor cells detachment,tumor cells transfer and invade the surrounding tissues,tumor cells circulate with the blood or lymph circulation and survival in them,the interaction of the tumor vascular endothelial growth factor and cells and interaction in the target sites,tumor blood vessels are worn out by the tumor cells,the formation of secondary tumors in the material organs and invade to the surrounding tissues,spreading throughout the body.Thus,the majorly late peried of the malignant tumors is transference.Colorectal cancer is one of the prevalent tumors in the digestive tract tumor.P53-induced gene 3(PIG3)is one of the downstream genes of p53 and participates in DNA damaging and apoptosis.It is testify that the expression of PIG3 can be found in many tumors,and connected to the tumors biological behavior.Currently,reports that the influence of PIG3 on colon cancer cell invasion and metastasis and related mechanism has not been found..AimsTo investigate the effect of PIG3 gene on invasion of colon cancer cells,providing the laboratory evidence for the feasibility of PIG3 gene used to cure the colorectal cancer.Methods(1)Colorectal cancer tissues assay:Using immunohistochemical method and qRT-PCR method to examine the expression levels of PIG3 in the colorectal cancers and normal tissues,analyzing the relationship between the expression levels of PIG3 and clinic pathological features of colorectal cancers.(2)SW480 Cell assay:(1)Groups:according to the immunohistochemical and qRT-PCR results,SW480 cell within pBK-PIG3 plasmid is the experimental group,SW480 cell without pBK-PIG3 plasmid is the control group.(2)Detecting the expression levels of PIG3 in two groups by Western blotting test and qRT-PCR method.(3)Analysising the invalid ability of PIG3 in SW480 cell through the transwell experiment and wound hilling experiment.Results(1)Colorectal cancer tissues assay results:Compared to the normal tissues,the expression level of PIG3 is very low which relates to the differentiation of the colorectal cancers tissues,the higher differentiate degree,the higher expression levels;qRT-PCR result resumes that the expression level of PIG3 in the colorectal cancer tissues is lower than that in the normal colorectal tissues.(2)SW480 Cell assay results:(1)Western blot test and qRT-PCR method showed that the expression level of the experiment group are higher than that in the control group on protein and mRNA levels.(2)Transwell experiments showed that both the two groups have been transfected through the cell basement membrane after 24 h,but the number of transfected cells are significantly lower than that of non-transfected cells.(3)Wound hilling result shows that both of two groups have a cell floating after 24 h,but the degree of the experiment group is lower than that in the control group.Conclusions(1)PIG3 expressed both in the normal colorectal tissues and colorectal cancer tissues,and the expression level of PIG3 in normal colorectal tissues is higher than that in colorectal cancer tissues.(2)The high expression level of PIG3 in SW480 cell can suppress the invalid ability of the tumour. |
