The TCM of Radix Phytolaccae is the dry root of Phytolacca acinosa Roxb.and Phytolacca americana L.in Phytolaccaceae.It is bitter in flavor,cold in nature,and belonging to the lungs,kidneys,large intestine.It can smoothen secretion,remove swelling and lump.Also,it has immunoregulatory and anti-inflammatory effects in modern pharmacological studies.Through the research of literatures,it is recorded that Phytolacca acinosa has the curative effects for nephropathy.However,it is toxic,and has been reported in clinical application.In order to study the effect on the treatment of nephropathy and the difference of toxicity from crude and vinegar processed Phytolacca acinosa,as well as the main material from them.This experiment carried on the crude and vinegar processed Phytolacca acinosa of nephropathy treatment effect and toxicity research,then analyzed the main different compounds between them.We choose the adriamycin nephrosis(AN)rats model to explore the difference of the nephropathy treatment effect between crude and vinegar processed Phytolacca acinosa by ight microscope,immunohistochemical methods,and so on.The results show that both crude and vinegar processed Phytolacca acinosa can reduce the urinaty protein from AN rats in 24 hours,reduce the serum CHO content,and it has significant differences between crude and vinegar processed Phytolacca acinosa.Both of them can reduce Urea and Cre level;The vinegar processed Phytolacca acinosa can significantly callback the Alb and TP level and reduce the kidney exponent.The two of Phytolacca acinosa can influence the expression of immune inflammatory factor,such as the expression of IL-1β and IL-2 level,reduce the renal NO content.They can serum MDA content to improve the antioxidant function of AN rats.The renal tubular proteinuria has reduced under the light microscope after HE staining.This research shows that both crude and vinegar processed Phytolacca acinosa can treat the AN rats.In addition,vinegar processed Phytolacca acinosa can enhance the curative effect in AN rats.To explore whether the difference of curative effect is caused by the toxicity of Phytolacca acinosa,we explored the acute and chronic toxicity of them.The results show that the water decoction of crude and vinegar processed Phytolacca acinosa found no obvious toxicity by giving mice.The chronica oral toxicity experiment shows that high dose of crude Phytolacca acinosa can cause Alb and TP level significantly reduced,the MDA and SOD level significantly lower after 4 weeks.It is visible that the injury of renal tissue injury under the light microscope.Experiments show that the crude Phytolacca acinosa can lead to the damage of kidney,and vinegar processed Phytolacca acinosa has a lower toxicity.Through the above study,it is known that crude and vinegar processed Phytolacca acinosa has different degrees of curative effects of nephropathy and toxic.To explore the main chemical composition from them,the experiment using UPLC/Q-TOF MS system with 0.1%formic acid water-0.1%formic acid acetonitrile as mobile phase,column temperature is 40℃,the flow rate of 0.4 mL/min,the injection volume is 2 μL,adopts the ESI ion source for data collection in the positive ion mode by ACQUITY UPLC BEH C18(1.7 μm,2.1 mm×100 mm)Waters column.Using principal component analysis(PCA)and orthogonal partial further squares discriminant analysis(OPLS-DA)to analyse and identificate the main compounds which cause the the difference between crude and vinegar processed Phytolacca acinosa.We Select and identify five compounds,they are triterpenoid sapogenin of 2-hydroxy elculentic acid,elculentic dicarboxylic acids A,Phytolaccagenin and esculentic acid G.And an other types of compounds is y-aminobutyric acid.The results provide a reference basis for the pharmacology and toxicity changes between crude and vinegar processed Phytolacca acinosa.This article explores the pharmacology and chemical experiments to explain the view of“attenuated synergies”by stir-frying with vinegar.Analysis and identify five compound who may cause the differences between crude and vinegar processed Phytolacca acinosa.These experiments can provided the scientific basis of the principle of Phytolacca acinosa,in order to guide the clinical medication of Phytolacca acinosa. |
