The Screening Of The Effective Components Of Gentianella Acuta Which Treats Alcoholic Liver Injury In Rats

Gentianella acuta which is also called "ku long dan" widely spreads in the north east of China,Inner Mongolia,Shan Dong,Shan Xi.Gentian ella acuta which possesses abilities to treat icterohepatitis,stenocardia,tr auma and infection,etc.has an effect of clearing heat and promoting diur esis,relieving the jaundice symptom.Gentianella acuta can cure and prev ent ourselves from cardiovascular disease when it has been drunk as te e for a longtime.Gentianella acuta contains xanthones,favonoids,triterpe noids,iridoids,lignans,steroids however there is seldom reportsabout the pharmacology and chemistry.Alcohol abuse has been a serious public health poblem.Alcohol liver disease mainly contains alcoholic fat liver,alcoholic hepatitis,alcoholic liver fibrosis and alcoholic cirrhosis.Spirits culture has a long history i n china however the name of alcoholic liver disease can not be traced from the traditional chinese medicine.But the diesese resulting from alc ohol can be found in the ancient chinese medical book.Herbal medicine is made up of many complex compounds.Nowadays,more and more scientists focus on the active constituents of herbal me dicine.This experiment is mainly about the active part of gentianella ac uta which can resist alcoholic liver disease.First,gentianella acuta was extracted by 70%、95%alcoholsolvent,th en it was enriched by AB-8 macroporous resin.Finally we got five parts extracts:water elution part,30%,50%,70%,95%etanol elution part of ma croporous resin.We screended the effective parts by pharmacodynmic m ethods.The study used intragastric administration to replace the model o f alcohol-induced liver injury in rats.Compared with the ethanol extract,we screened the active parts from the five etanol-water elution parts by contrastingtest results such as ALT,AST,TG,ALB,TP,MDA,SOD,GSH.The phacmacodynamics research results showed that the model of hepatic i njury was established by intragastrically administered with gradient alco hol for eight weeks.The group of gentianella and,70%etanol elution partsreduced the levels of serum ALT,AST,TG,ALB significantly.(P<0.05)At the same time the three groups abovecan reduce the levels of M DA in liver tissues while the three groups can promote the contents of SOD,GSH in liver tissues.(P<0.05)In the model group most structures of hepaatic lobules were destroyed by the liver histopathological obser vation.All of the results above indicaed that gentianella acuta can protect the rats from alcohol liver injury,and the effective parts of this herbal medicine is 70%etanol elution parts.
