Belonging to the family Goodeniacea,Scaevola Sericea(synonyms: Scaevola koenigii;Scaevola taccada;Scaevola frutecens)is a salt-water shrub,having good biological activities and a variety of compounds.Because the biological activities and chemical composition of Scaevola Sericea in Guangxi Fangchenggang had not been reported before,they were studied in the paper in order to develop the mangrove plants of Guangxi.There are several aspects elaborated as following.Chapter one: Preface.Firstly,we introduced the ecologic characteristics,the geographical distribution and botanical classification of Scaevola sericea briefly.Then the reseach status at home and abroad was reviewed including the folk medicine,bioactivities and chemicals.Lastly,the topic of the subject was put forwared.Chapter two: The study on bioactivity of Scaevola sericea.Prior to the extraction,the leaves,barks and roots were shade dried in order to prevent photolysis and thermal degradation,chopped into small pieces,ground coarsely to powder form in a mechanical grinder and immersed in 95% ethanol.The extracts were further concentracted by recovering excess solvents to a thick oily natured crude in a rotary evaporator at reduced pressure.The thick oily crude were extracted again with various solvent like petroleum ether,ethyl acetste,n-butanol and water.Likely,the extracts were further concentracted to thick oily crude for studies such as antitumor,antioxidation,antibacterial activities and so on.Results as follows: 1.The crude of Scaevola sericea has good antitumor activity.The IC50 is below4.5 μg/mL against Hela cells for the bark crude extraction;The IC50 are 7 μg/mL aginst 7901 cells and 23 μg/mL against 7721 cells for the petroleum ether extraction of bark;The IC50 are 31μg/mL aginst 7404 cells and 40μg/mL against 7721 cells for the ethyl acetate extraction of leaf.Further more,all the crude extraction are normal role on the growth of 293 T cells.2.The antioxidant activity of ethyl acetate in stem and leaf of Scaevola sericea is better than BHT.3.The Tyrosinase inhibition activity of n-butanol extraction of bark is close to the rutin.4.All extraction has antibacterial activity.Among the fractions tested for antibacterial activity against Staphylococcus aureus,the ethyl acetate extract of bark was found to be active with MIC at 250μg/mL.Besides,some compounds also has antibacterial activity and good antitumor activity.Compound 5,3,4,5,8 and 9 have good antitumor activity against HepG2 cells with IC50 at 15 μg/mL,32 μg/mL,47 μg/mL,56 μg/mL,15 μg/mL and 8 μg/mL.Compound 9 has antitumor activity against Hela cells with IC50 at 38 μg/mL.Meanwhile,all the compounds play a normol role on 293 T cells.Chapter three: The analysis of chemical composition of Scaevola sericea.The inorganic element was analysed by ICP-MS and ICP-AES.Elements which are beneficial for the body including Sr,Mo,P,Fe,Ca,Cu,Cr and Mg are found in Scaevola sericea.Element Na,K,Ca were rich in Scaevola sericea of which the content of element Na in leaf was three time more than in root.Besides,the main volatile chemical constituents of petroleum ether and ethyl acetate extraction of Scaevola sericea leaves and stem were analyzed by using gas chromatography-mass spectrometry(GC-MS),including linoleic acid,palmitic acid,phytol,linoleic acid ethy ester,ethyl linolenate and so on.Chapter four: The isolation and structure identification of Scaevola sericea.In this research,eleven compounds have been isolated from Scaevola sericea.Five of the isolated compounds from the petroleum ether extraction of bark are known—stigmasterol,epifriedlanol,α-taraxerol,friedelin and γ-Amyrin acetate.Two of the isolated compounds from the ethyl acetate extract of bark are myricadiol and enol polymer.Three of the isolated compounds from n-butanol extraction of bark are also known--laciniatoside Ⅴ,ursolic acid-β-D-Glucopyranosyl ester and pomolic acid-β-D-Glucopyranosyl ester.Also a large quantity of inorganic salt was found in water extract of leaves and analysed by ICP-MS and ICP-AES.The result shows that element Na,Mn,K are rich in inorganic salt and the mass fraction are 28.395%,29.51% and 7.38%. |
