Objective:Based on the observation of the subject and make the theory of Jiajiansa-njiasan curative effect for the treatment of carbon tetrachloride induced liver fibrosis rat model,study the effect on the expression of microRNAs in rat model,explore the molecular mechanism of anti liver fibrosis of the party from the gene level.Methods:30 male Wistar rats were randomly divided into normal group,model group and Jiajiansanjiasan group,with each group containing only 10 rats.After routine fee-ding of 7days,the model group and the addition and subtraction group were injected subcutaneously with CCl4 to replicate the animal model of hepatic fibrosis.Addition and subtraction of the three groups of CCl4 from the date of subcutaneous injection,that is,given the appropriate drug gavage,the amount of the drugs were given by the0.1mL/100g body weight of the corresponding drugs,1 times a day.Normal group and model group were given the same volume of distilled water,1 times a day.A total of 8 weeks.After the treatment of 56days,the rats were killed by anesthesia.Each rat was cut two pieces of liver tissue,and was used for HE and MASSON staining to observe the pathological morphology and microRNA gene sequencing.Results:1.The control group rats’hepatic lobulestructure were integrated,without liver ce-ll degeneration or necrosis,without fibrous connective tissue hyperplasia.The model gr-oup’s rats appeared cellular swelling,ballooning degeneration,lipoid degeneration,lar-ge scale of fibrous tissue proliferation from central veins to portal area,formation ofpseudolobuli.Jiajiansanjiasan group rats also appeared cellular swelling,small scale of fibrous tissue proliferation around portal area.Compared with control group,the liver tissue inflammation activity grading/hepatic fibrosis staging and collagen fiber areapercentage increased significantly in model group and Jiajiansanjiasan group rats.Compared with model group,the liver tissue inflammation activitygrading/hepatic fibrosis staging and collagen fiber area percentage decreased significantly in Jiajiansanjiasan group rats.2.The difference of fold(two change>1)and the significant level(P<0.05)were evaluated at different levels,and the differences of microRNAs were screened.In the model group compared with normal group,there are 37 different microRNAs,of which 29 were up-regulated and 8 were down-regulated;Jiajiansanjiasan group compared with normal group,there are 100 different microRNAs,of which 80 were up-regulated and 20 were down-regulated;at the same time,compared with the model group and normal group compared with the model group and selected 14different expressioned microRNAs.Conclusions:1.CCl4 subcutaneous injections can successfully replicate rat immunity liver fibrosis model.2.Jiajiansanjiasan can improve pathological morphology,partserum liver fibrosis markers.3.Using high-throughput microRNA sequencing technology were detected and screened some differentially expressed microRNAs,they may give part expression to scientific connotation of“Zhukejiao”theory by the interaction with each other.4.Jiajiansanjiasan may play an important role in liver fibrosis through the regulation of miR-146b-5p,miR-125b-5p and miR-200b-3p regulation of the corresponding function or pathway. |
