Study On The Influence Of Tonifing Deficiency And Resolving Stasis On MRNA Expression Of BDNF And Its Receptor TrkB In The HyPothalamus Of Post Partum DePression Model Rats | Posted on:2019-01-22 | Degree:Master | Type:Thesis | Country:China | Candidate:M Liu | Full Text:PDF | GTID:2394330566994941 | Subject:TCM gynecology | Abstract/Summary: | PDF Full Text Request | Objective:In this PaPer,the rat models of PostPartum dePression(PPD)were studied as the research vector,and the Chinese medicine chanshu granules of reinforcing deficiency and removing blood stasis was involved in the intervention,to observe the behavioral changes,the level of serum estradiol(E2),to detect the exPression levels of neurotroPhic factor(BDNF)and tyrosine kinase rece Ptor(TrkB)mRNA in the hyPothalamus of PPD model rats.In order to exPlain the regulation and amelioration effect of the thera Peutic method of reinforcing deficiency and removing blood stasis and the Chinese medicine chanshu granules on PPD model rats.Combined with the Previous exPerimental results of this study,we exPlore the Possible role and influence of BDNF and its selective recePtor TrkB in different Parts of PPD model rats’ Brain tissue including HyPothalamus,frontal lobe,hiPPocamPus.And then study on the mechanism of reinforcing deficiency and removing blood stasis on PostPartum dePression furtherly.Methods:1.According to the scores of the oPen field,the mature female SD rats we re randomly divided into normal control grou P(Ctl grou P)and PPD model gr ou P(PPD grouP).The mature female SD rats were ovariectomized bilaterally and received subcut-aneous injection of hormone(estrogen Progestin)for the mo del of PostPartum de Pression.Then all of the Successful PPD rats were rando mly divided into 6 grouPs: The shenguiren mixture of high dose grouP(S-H g rouP),the shenguiren mixture of medium dose grou P(S-M grou P),the shenguir en mixture oflow dose grou P(S-L grouP),the estradiol benzoate grouP(EB grou P),sertraline hydrochloride grou P(Ser grouP),and the model grouP(Mdl grouP)..The S-H grouP,S-M grouP,S-L grouP and Ser grou P were Pre Pared with the corresPonding drugs administration by gavage resPectively once a dayfor 4 weeks.The Ctl grou P and Mdl grouP were Pre Pared with Isobaric Pur e water administration by gavage.Using sucrose water consumPtion test,oPen field test and forced swimming test to conduct behavioral tests on all animals after modeling.2.After 4 weeks of intervention,the serum estradiol level was detected by radioimmunoassay(RIA).The ex Pression of BDNF and TrkB mRNA in hy Poth alamus of rats was detected by real-time quantitative PCR(RT-PCR).Results:1.Behavioral index results:The data after drug intervention showed that the behavioral indexes of Mdl grouP exerted a tendency to de Pression comPared with the Ctl grouP,and the indexes after administration for 4 weeks were slightly imProved comPared with the Mdl grouP.After administration for 2 weeks,the sugar consumPtion of the S-M grouP and EB grouP was significantly increased than that of the Mdl grouP(P < 0.001),and that of the S-H grou P,S-M grouP,S-L grouP and EB grouP were significantly higher than the control grou P(P < 0.001).After administration for 2 and 4 weeks,the scores of oPen field ex Periment level in each grouP were significantly higher than those in the Mdl grouP.(P < 0.001).The immobility time of swimming in all grouPs excePt Ser grouP was decreased(P <0.001,P < 0.01).2.The level of serum E2 in modle grouP was lower than that in Ctl grouP(P >0.05).The levels of serum E2 in EB grouP and S-M grouP were significantly higher than that in Mdl grouP.3.RT-PCR results: ComPared with Ctl grouP,the exPression of BDNF and TRKB mRNA in hyPothalamus of rats in Mdl grou P were significantly down-regulated(P < 0.01 and P < 0.05).ComPared with Mdl grouP,the ex Pression of BDNF mRNA in EB grouP and S-M grouP were uP-regulated(P < 0.05).There was no significant difference in the exPression of TrkBmRNA in different dose grouPs of Shenguiren mixture(P > 0.05).Conclusion:1.Shengui Ren mixture can im Prove the dePressive behavior of PPD model rats.2.Shengui Ren mixture can uPregulate the level of serum E2 and imProve the low state of serum E2 in PPD model rats,which further Proves that Shengui Ren mixture has estrogen-like effect,suggesting that Shengui Ren mixture may Play an antidePressant effect through estrogen mediated.3.Shengui Ren mixture tonifying deficiency and removing blood stasis may Play an antidePressant role by regulating the interaction between estrogen and hyPothalamic neuroendocrine system,imProving the exPression of BDNF mRNA and trkb mRNA exPression in hyPothalamus.can increase the exPression level of BDNF mRNA in hyPothalamus. | Keywords/Search Tags: | tonify deficiency and remove blood stasis, PostPartum dePression, hyPothalamus, Brain derived neurotroPhic factor, TroPomyosin recePtor kinase B | PDF Full Text Request | Related items |
