| Background and objective: Esophageal cancer is a malignant tumor,which is originally found in the epithelium of the esophagus.It is one of the common malignant tumors,and it’s seriously threatening humanity’s health.Esophageal cancer is sixth of the causes of cancer death around the world.Meanwhile,it’s also a high incidence area of esophageal cancer in our country,especially in Lin County in Henan,Ci County in Hebei and Yangcheng in Shanxi.And the incidence and mortality rate of esophageal cancer are still very high in China.Radiotherapy and surgery are the most important treatment for esophageal cancer,and radiotherapy is the most effective treatment in the middle and upper segment of the esophagus.But because the effect of radiation therapy is affected by many factors,it is important to find the drugs that can be sensitized.In recent years,traditional Chinese medicine as a radiosensitizing drug has become a hot topic in radiobiological research at home and abroad.Curcumin is an acidic polyphenol extracted from herbs,which has a wide range of pharmacological effects,and can regulate a variety of signal transduction molecules,such as chemokines,transcription factors,cell cycle regulatory proteins,etc.Curcumin has anti-inflammatory,anti-coagulant,anti-oxidation and other effects due to its molecular biological characteristics.And the radiosensitivity and its possible mechanism on human esophageal cancer cell Eca-109 by curcumin are investigated in this paper.Methods: 1.Different concentrations of curcumin were placed for 24 and 48 h,such as,0μmol/L,5μmol/L,10μmol/L,20μmol/L,40μmol/L,80μmol/L,and measured the cell proliferation by CCK-8.And the OD values were detected by Microplate Reader after adding the CCK-8 reagent.2.The cloning experiment was conducted to determine the esophageal cancer cell of radiotherapy sensitization by curcumin.The experiments were divided into the experimental group with curcumin treatment and the control group without drug.The two groups were treated with different rays,such as,0Gy,2Gy,4Gy,6Gy,8Gy,and cultured for 2-3 weeks.After that,the difference of the clone formation rate of the Eca-109 cell line of the esophageal cancer cells was measured,and the SER value was calculated.And the single target multi hit model is selected to fit the survival curve,using GraphPad Prism 5.3.The apoptosis rate of curcumin in esophageal cancer cell line Eca-109 was detected by flow cytometer.The experimental group and the control group were irradiated with different radiation,such as,0Gy,2Gy,4Gy,6Gy,8Gy.After culture 24 h,cell and cell culture medium were collected.And the apoptosis rate of esophageal cancer cells handled by curcumin was detected by flow cytometer.4.The cell cycle of esophageal cancer cell line Eca-109 was detected by flow cytometer.The control group and the experimental group were irradiated with radiation dose 4Gy and cultured for 8h.And the cell cycle for esophageal cancer cells handled by curcumin was detected by flow cytometry instrument.Results:1.Curcumin has significant inhibitory effect on human esophageal cancer Eca-109 cells,and there is a dose and time dependence(P<0.05).2.There was a significant difference in the clone formation rate of the Eca-109 cell line of esophageal cancer cells irradiated by different rays(p<0.05)after adding the curcumin.It was proved that curcumin had sensitization to Eca-109 of esophageal cancer cells.And the radiotherapy sensitization ratio was measured 1.37 by plate colony formation assay.3.Flow cytometry instrument was used to detect the apoptosis rate of Eca-109 cells treated with curcumin and there was a statistical difference(P<0.05).4.It’s showed that the cell cycle of esophageal cancer cell line Eca-109 treated by curcumin was statistically different from that of pure irradiation,and the cell cycle was delayed in G2/M period.Conclusion: Curcumin that has been proved that there is radiotherapy sensitization on human esophageal cancer cell line Eca-109,which maybe caused by blocked of cell cycle. |
