| Objective To explore the interaction between 5-hydroxytryptamine(serotonin)receptor 1B(5-HTR1B)gene and serotonin transporter(5-HTT)gene on the 5-hydroxytryptamine system,and its relationship with attempted suicide and non-suicide self-injury in youth.Methods The iMLDRTM multiplex SNP typing technique was used to examine one locus(rs6296)of 5-HTR1B gene,and three loci(rs1042173,rs140701 and rs20667134)of 5-HTT gene in the attempted suicide group,non-suicide self-injury group and the control group.Among them,there were 70 youth with attempted suicide involved in the attempted suicide group,55 youth with non-suicide self-injury in the non-suicide self-injury group,and 112 healthy youth subjects in the control group.Furthermore,analysis of gene-gene interaction was achieved by generalized multifactor dimensionality reduction,(GMDR).Results Between attempted suicide and controls,there was not significant difference in the genotype frequency distribution and allelic frequencies distribution of 5-HTT rs1042173,rs140701,rs2066713(P>0.05).There was not significant difference in allelic frequency distribution of 5-HTR1B rs6296 in attempted suicide group and control group(P>0.05).There was significant difference in the genotype frequency distribution of 5-HTR1B rs6296 between the attempted suicide group(CC31.4%、CG 31.4%、GG 37.1%)and the control group(CC 19.8%、CG53.2%、GG 27%)(χ~2=8.327,P=0.016),however,no statistical difference was found after Bonferroni correction(P>0.05).Between non-suicide self-injury and controls,there was not significant difference in the genotype frequency distribution and allelic frequencies distribution of 5-HTR1B rs6296 and 5-HTT rs1042173,rs140701,rs2066713(P>0.05).Between attempted suicide and controls,GMDR analysis showed that the interaction between 5-HTT and 5-HTR1B gene existed in the two-site model(rs1042173、rs6296)(test accuracy:0.6185,cross validation consistency:10/10,P=0.0210~0.0220),three-site model(rs1042173、rs140701、rs6296)(test accuracy:0.6445,cross validation consistency:10/10,P=0.0030~0.0040)and four-site model(rs1042173、rs140701、rs2066713、rs6296)(testaccuracy:0.6120,cross validation consistency:10/10,P=0.0320~0.0330).Between non-suicide self-injury and controls,GMDR analysis showed no interaction between 5-HTR1B rs6296 and 5-HTT rs1042173,rs140701,rs2066713(P>0.05).Conclusion There is interaction between 5-HTR1B gene and 5-HTT gene in attempted suicide,and it may be associated with attempted suicide in youth.There is not interaction between 5-HTR1B gene and 5-HTT gene in non-suicide self-injury,and it may be not associated with non-suicide self-injury in youth. |
