Objective: By detecting the rats model of Chronic Atrophic Gastritis(CAG)in general conditions,gastric mucosa tissue pathological change,expression level of change in Hippo/TAZ signaling pathway and its related proteins,including TAZ,LATS2,MST1 in gastric mucosa tissues,we can explore the mechanism of HuaZhuoJie Du formula in CAG treatment,further perfect ZhuoDu theory system,provide objective basis and new ideas for clinical prevention and treatment of the CAG.Methods: 70 healthy adult male SD rats were divided into Normal Group(N)12 and Model Group(M)58.After the success of using combined modeling method to establish rats model,we randomly divide them into 5Groups: Spontaneous Recovery Group,High-doze Group,Moderate-doze Group,Low-dose Group,Vitacoenzyme Group,each group of 10.The HG Group,MG Group and LG Group were given different concentrations of HuaZhuoJieDu solution,and the VG Group was provided with the enzyme suspension.Using HE staining to observe the gastric mucosa tissue pathological changes,Immunohistochemical method(SP),Western Blot and Real-time quantitative Polymerase Chain Reaction to observe expression level changes of protein TAZ,LATS2,MST1 in gastric mucosa tissues.SPSS 21.0was used to analyze all data.Results:1.The general condition In building stage,the rats in Group N were generally in good condition.The rats in Group M were not in good condition and had obvious changes.In the treatment stage,the symptoms of rats in HG Group improved significantly.The general conditions of the rats in MG and VG Group were partly improved,and the general improvement in LG Groupand SR Group was not obvious.2.Effect of HuaZhuoJieDu formula on TAZ expression in gastric mucosa of CAG rats.HG Group’s average optical density,the last grey value,mRNA were below the VG Group,MG Group,LG Group,the difference was statistically significant(P<0.05),MG Group and VG Group,there was no statistically significant difference(P=0.237,P>0.05),LG Group is higher than the VG Group,the difference was statistically significant(P<0.05).The MG Group was lower than LG Group,and the difference was statistically significant(P<0.05).3.Effect of Hua ZhuoJieDu formula on LATS2 expression in gastric mucosa of CAG rats.HG Group’s average optical density,the last grey value,mRNA were higher than in VG Group,MG Group,LG Group,the difference was statistically significant(P<0.05).MG Group,LG Group is lower than the VG Group,the difference was statistically significant(P<0.05),and the MG Group was higher than LG Group,the difference was statistically significant(P<0.05).4.Effect of HuaZhuoJieDu formula on MST1 expression in gastric mucosa of CAG rats.HG Group’s average optical density,the last grey value,mRNA were higher than in VG Group,MG Group,LG Group,the difference was statistically significant(P<0.05).MG Group,LG Group is lower than the VG Group,the difference was statistically significant(P<0.05),and the MG Group was higher than LG Group,the difference was statistically significant(P<0.05).Conclusions:HuaZhuoJieDu formula can reduce the expression level of TAZ protein and increase the expression level of LATS2 and MST1 protein in gastric mucosal tissue of CAG rat model.High dose HuaZhuoJieDu formula can improve the pathological condition of gastric mucosa significantly.In the regulation process of Hippo/TAZ pathway,the negative regulation of TAZ protein and positive regulation of both LATS2 and MST1 were superior to the Vitacoenzyme Group.
