Purpose: To observe the neuroprotective effect of Zhongfeng Gao(ZFG)on cerebral ischemia injury in rats,and explore the mechanism of this protective effect.Methods: 100 SPF-grade male SD rats are randomly divided into normal group,sham operation group,model 1d,3d,7d,14 d group,stroke paste 1d,3d,7d,14 d group,each group of 10.And after the success of mold building,the drug is administered to the rats’ stomach in the next day.Then after 1d,3d,7d and 14 d rats are given the drug,the rats are put to death in batches,collected and become fixed specimens.Next,observe the neurological function score of MCAO rats,measure the volume of cerebral infarction of rats at different time by TTC staining method,test ischemic brain hippocampus,the NGF,Notch1,and the hes1 gene expression by RT-PCR,and detect the expression of NGF protein in the hippocampal tissue of ischemic brain by Western blot.Results:1.Comparison of neurological function scores in rats.After the ischemia-reperfusion injury,the rats in the model group and the apoplexy group all appeared nerve function defect after operation,and then 1d,3d,7d to 14 d,the score gradually decreased.The neurological function of the two groups recovered with time,and the score decreased gradually,but the decrease of the ZFG group was significantly faster than that of the model group.The scores of 7d and 14 d were significantly worse than those in the model group.Difference(P<0.05).2.TTC staining method for testing the cerebral infarction volume of rats.Compared with the sham operation group,the infarction rates of all groups are significantly increased except for the blank group,and the difference has statistical significance(P<0.01).Compared with the 3-day group of the model,the infarct volume in the 3-day group of ZFG is reduced,but compared with the model 7 days,the infarct area decreases in the 7-day group of ZFG,and the difference has statistical significance(P< 0.05).3.RT-PCR method to detect the gene expression in the hippocampus of the hippocampus.(1)Results of Nerve growth factor(NGF): compared with the blank group and model group: NGF gene has different levels of expression in brain tissue.Compared with the blank group,the expression of NGF in the model group and the ZFG group significantly increased,and the difference is statistically significant.(P<0.01).Compared with the sham operation group,the difference between the model group and the ZFG group was statistically significant.(P<0.05).Compared with the model group,there was no significant difference between the 1D group and the 3D group at the same time(P>0.05).The NGF gene was obviously expressed in the 7d group of ZFG in the same time,and the difference was statistically significant(P<0.01).The difference of ZFG for the same time in 14 days group was statistically significant(P<0.05).(2)Result of Notch1 :The expression of Notch1 gene in model group and ZFG group was significantly higher than that in blank group and sham operation group,the difference is statistically significant.(P<0.05);And the Notch gene in ZFG group is higher than that in the model group of 1d,3d and 7d at the same time,the difference is statistically significant.(P<0.05).Then compared with the model 14 d group,the expression of Notch gene increased significantly in the ZFG 14 d group,and the difference is statistically significant(P<0.01).(3)Results of hes1: the hes1 gene of brain tissue in model group and ZFG group was significantly higher than that in blank group and sham operation group,and the difference is statistically significant(P<0.01;P<0.05)However,there was no significant difference between the model group and the model group on the 1st and 3rd day after ZFG and the difference was not statistically significant(P>0.05).Compared with model on 7 and 14 days,the expression of hes1 gene in the ZFG group was significantly higher than that in the model group at the same time point.and the difference is statistically significant(P<0.01).4.Detection of NGF protein expression in hippocampus of rats with cerebral ischemia by Western blotThe expression of NGF protein in the model group and ZFG group was significantly higher than that in the blank group and the model group,and the difference is statistically significant(P<0.01).But compared with the model group at the same time,there was no significant difference between the 1 d group and the model group(P>0.05);compared with the model 7d group,the difference at the same time in ZFG group was statistically significant(P<0.01).Compared with the model for 3 days and 14 days,the difference at the same time in ZFG group was statistically significant(P<0.05).Conclusions:1.ZFG can improve the symptoms of nerve dysfunction in MCAO rats.This effect was not obvious in 1-3days after infarction,but was the most obvious at 7days,and still was protective after 14 days.2.ZFG is used to treat cerebral infarction and the recover nerve function,which could be achieved through Notch signaling pathway. |
