Objective:Obesity is a chronic metabolic disease that is closely related to the chronic inflammation of fat.The imbalance of polarized M1/M2 macrophages in adipose tissue is crucial in the chronic inflammatory response caused by obesity.Therefore,based on the classic food-biased animal model,this article explored the benign regulation of electroporation on the polarization balance of adipose tissue M1/M2 macrophages in obese mice,and provided a scientific basis for the prevention and treatment of obesity by Chinese medicine.Methods:C57 mice were fed for 1 week and fed a high-fat diet for 12 weeks.Obese mice were randomly divided into model group(M),electroacupuncture 7 days(A7)group,14 days(A14)group,21 In the day(A21)group and the 28-day(A28)group,7 mice in each group were randomly selected 7 as failed fatty rat(L)groups.The treatment group received acupuncture at Tianshu,Guanyuan,Zusanli,and Sanyinjiao.After the gas was treated,the electroacupuncture apparatus was switched on.The other groups were fixed by the same method but were not treated.Observe the different time windows and observe the animal body weight,spleen mass,fat mass and morphological characteristics around the epididymis in each group.The contents of IL-4,TNF-α and IL-10 in spleen,IL-4,TNF-α,IL-10,leptin and chemerin in the epididymis were determined by ELISA.The gene expression of F4/80,CD11 c,iNOS,CD206 and endogenous peroxidase(CAT)was detected by fluorescence quantitative PCR.Results:The body weight(40.61±6.59),epididymal fat mass and cell diameter of the simple obesity group were significantly higher than those of the C group(24.64±3.69)and the L group(29.44±2.33)(P<0.01);after electroacupuncture treatment,each treatment The fat mass and cell diameter of the epididymis decreased significantly inthe group,among which,the body weight of the A28-day group(28.68±4.12)had the most obvious effect(P<0.05).There was no significant difference in spleen mass between groups(P>0.05).After electroacupuncture treatment of obesity,the levels of pro-inflammatory cytokines TNF-α and anti-inflammatory cytokines IL-4 and IL-10 were significantly increased(P<0.01)in liposuction and epididymal fat around each group;The levels of chemerin and chemerin decreased significantly(P<0.01).Compared with group A7,the levels of A21 group and A28 group decreased significantly(P<0.05).After treatment with electroacupuncture in obese mice,the expression level of F4/80 on the surface of macrophages in adipose tissue was increased,and the expression levels of CD11 c and iNOS on the surface of M1 macrophages were significantly reduced,and the expression of M2 macrophages was decreased.The gene expression of surface expression marker CD206 was significantly increased(P<0.01),and the expression of CAT gene was increased(P<0.05).Conclusion:Electroacupuncture can significantly improve the obese state of mice,and through the benign regulation of M1/M2 macrophage polarization ratio imbalance and dysfunction,adjust the fat immune dysfunction,reduce the fat chronic inflammatory response process.
