| Objective: The expression and change of HES1,5 and HERP1,2 and other related genes in rabbits with steroid induced osteonecrosis of the femoral head(SANFH)before and after modeling,at different stages of modeling and after modeling were observed by observing Huoxue Tongluo decoction.By means of light microscopy and electron microscopy,we can observe the change of rabbit femoral head bone tissue specimens from a pathological point of view,and study the possible mechanism of Huoxue Tongluo Decoction in preventing and treating SANFH.It will provide a new treatment for this disease.Methods: 192 Japanese big ear rabbits were randomly divided into four groups: A,B,C and D.They were the prevention and treatment group,the treatment group,the model control group and the blank group,each group was 48.A,B,C three groups in third weeks when using he Xijing modeling method of manufacturing model of osteonecrosis of the femoral head,molding cycle time for 6 weeks.A group in the second week(a week before modeling)began to feed medicine food made of Huoxue Tongluo Decoction;B rats in groups and fed with Chinese herb feed;C and D two groups with the same.On the second,fifth,eighth weekend each specific time node of each group were randomly selected 8 rabbits with PCR(real-time quantitative method)method for the determination of HES1 in serum,the content of HERP1,5,2,and the continuous and dynamic observation of the femoral head size,appearance and morphology,and double side interception of the femoral head for pathological sections,electron microscopic observation on the histomorphology of bone tissue by light microscope and electron microscope,the statistical rate of empty lacuna.Results:1,HES1: at each weekend,the expression values of group D were all smaller than those of group A,and the gap between the two groups gradually decreased over time.At second weeks,the difference between the two groups was the largest(P<0.01),and the difference was statistically significant.The gene expression value of group B(treatment group)was higher than that of group D at the end of the week,and the statistical significance was clear between the two groups at the end of the second,fifth week.The results of every weekend in group C were higher than that of group D(P<0.01),and the statistical significance was clear.Compared with the three groups of A,B and C,the results of A and B were lower than those in the C group,while the test results of the A group were lower than that of the B group.2,HES5: at the end of the second,fifth week,the gene expression value of group A was obviously higher than that of group D.At the end of the eighth week,it suddenly decreased significantly,and was lower than that of the D group.There was no statistical difference between the two groups(P>0.05).The results were compared between the three groups of A,B and C(P>0.05),and the difference was not statistically significant.3,HERP1: in the weekend,A,C group comparison,the test results were lower than the C group,especially at the fifth weekend,between the two groups(P<0.05)statistically significant.At the second,fifth weekend,the gene expression values of group B and group C were all higher than those of D group.At the end of fifth weeks,B group,D two group,C group and D two group were compared respectively(P<0.01),and the statistical significance was clear.At the end of the eighth week,between groups of B,D,C and D(P>0.05),the statistical significance was not clear.4,HERP2: at the second weekend,the detection results in group A were slightly higher than those in group D,and at the fifth,eighth weekend,the detection results decreased significantly than those in group D,and the statistical significance of P>0.05 was not clear.The results of group B and C were compared with group D at the end of the week,and the statistical significance of P>0.05 was not clear.Compared with the three groups of A,B and C,the statistical meaning of P>0.05 was not clear.Conclusion: 1、 The mechanism and Hes1 protein gene was positively related to avascular necrosis of the femoral head,and 1 hormones,Huoxue Tongluo Decoction can promote bone cell necrosis repair and reconstruction,to prevent the negative correlation mechanism and Hes1 protein gene of bone cell apoptosis;and their relationship with the Herp1 gene also cannot know,its specific role the mechanism remains to be further study;2、 2,the regulation of Hes5 and Herp2 protein gene is not related to the pathogenesis of steroid induced avascular necrosis of the femoral head and the mechanism of Huoxue Tongluo Decoction to promote the repair and reconstruction of necrotic bone cells and prevent the apoptosis of bone cells.It may be because the Hes5 and Herp2 protein genes do not participate in the regulation of bone cells,but they can not exclude the regulatory response to other cells.3、 3.We can conclude that Hes1 may be the target gene of the main regulation in the downstream,and it is the target of action.The expression of hormone can results in upregulation of Herp1 protein gene,differentiation,apoptosis and bone cells,caused by the pathological results of femoral head necrosis,and the traditional Chinese medicine Huoxue Tongluo Decoction can also pass through the signal pathway,plays the antagonist effect,reverse regulation,the down-regulation of Herp1 protein gene expression,and reduce the incidence of femoral the bone lacuna rate,showed that Huoxue Tongluo Decoction can promote bone cell necrosis repair and reconstruction,and prevent bone cell apoptosis,plays the role in the early stage of early intervention.4、 4,at present,many western medicine treatment methods for steroid induced osteonecrosis of the femoral head mainly focus on "treating the disease",though it can also achieve good curative effect,but it is not enough attention for the disease that may happen soon,so we should make early intervention.The experimental research method of this subject is entirely based on the idea of "disease prevention before treatment",which not only can cure this disease,but also can play an early preventive and therapeutic effect on the disease that is about to happen.It provides a new idea and method for the treatment of this disease. |
