Objective:Investigate the application and mechanism of urine-derived stem cell transplantation in the injury of mouse potocytes.Methods:96 male BALB/C mice were randomly divided into control group,model group,BMSCs+model group and USCs+model group.Adriamycin nephropathy mouse model were made by tail vein injection of adriamycin at a dose of 10.5mg/kg and where the control group were injected with the same dose of saline.The fourth,fifth,sixth weeks,BMSCs+model group and USCs+model group both have three times of the injection of BMSCs and USCs(1x10~6/100ul).The control group and model group were injected with the same of saline.Then there is the observation for 12 weeks of the weight,urine protein,urine protein/creatinine,renal pathological changes,Podocin(foot cell marker)expression in mice.Results:The control group:body weight of mice trends to grow faster while the model group,USCs group and BMSCs group,the growth trend were similar.Compared to the model group,BMSCs and USCs transplantation had an increase in weight,the difference was statistically significant.Compared to the control group,urine protein,urine protein/creatinine ratio from the second week increased obviously in model group,BMSCs group and USCs group while after the transportation of BMSCs and USCs,the number decreased.the pathological results showed that adriamycin after modeling,model group,BMSCs group and USCs group of mice podocyte foot process fusion,glomerular matrix increased,basement membrane thickening,mesangial cell proliferation,and increased with time gradually increased,BMSCs and USCs after transplantation,the pathological changes reduced and BMSCs and USCs had no difference between the two groups;Western Blot results showed that Podocin expression was significantly decreased in mice after modeling,BMSCs and USCs after transplantation,the expression of Podocin increased,and BMSCs and USCs two There was no significant difference in the group.Conclusions:USCs transplantation can protect podocyte,improve renal pathology,protect kidney,USCs infection and genetic disease after transplantation.
