| ObjectiveTo explore the effects of ghrelin on the apoptosis and fractakine(FKN)expression in human umbilical vein endothelial cells induced by high glucose.MethodsHUVECs were clutrued in EMEM/LG 5.5mmol/L)for 10 h,10-7mol/L ghrelin was added and incubated for 2h and then incubated with media with33.3mmol/L glucose for 48h.The apoptosis of HUVECs was evaluated by flow cytometry(FCM)and through Hoechst 33258 straining.Expression of FKN was detected by Q-RT-PCR.Protein level of FKN was determined by western blot.ResultsHigh glucose significantly induced an increase in apoptosis in HUVECs which was partly inhibited by ghrelin(P=0.000),though the apoptosis of HG+ghrelin still higher than normal control(P=0.000).Compare with normal control,HG enhanced the fractalkine expression at both protein(P=0.000)and mRNA(P=0.001)levels,and both the protein and mRNA levels of FKN were decreased in HG+ghrelin(P=0.000).ConclusionGhrelin reduces the apoptosis rate of HUVECs in high glucose environment,the mechanism may be related with ghrelin intervation in inhibiting the expression of FKN in HUVECs. |
