Study On The Mechanism Of Cold Syndrome Hypertension And The Therapeutic Effect Of Hanjingtang By Professor Li Shimao

Objective: The purpose of this experiment was to study the mechanism of blood pressure rise caused by cold and high salt diet and the therapeutic effect of hanjingtang,and to determine the indications of hanjingtang,so as to apply it more accurately in clinic.Methods: 24 male SD rats were randomly divided into control group,model group and hanjingtang group.The other two groups were given 8%sodium chloride diet except the control group.It was placed in a freezer with a temperature of(-15±2)℃ for 3 hours every day.The rest of the time was kept in the clean room with the control group,and the control group was given common feed.Blood pressure was measured three times a week by caudal artery in all rats.The time was fixed from 8 o’clock to 11 o’clock in the morning.After taking the average of 6 weeks,the rats were treated with the traditional Chinese medicine called hanjingtang.After two weeks of treatment,all the rats were killed by decapitation.The levels of T3,T4,TSH,TRH in plasma and SOD,MDA in serum were measured.The hypothalamic liquid nitrogen was rapidly frozen and stored in the refrigerator at-80 ℃ to detect the protein and gene expression of UCP4 in the hypothalamus.Results: Compared with the control group,the combined stimulation of cold and high salt diet could increase blood pressure(P<0.05),increase the level of plasma T3,T4,TSH,TRH(P<0.05),decrease the content of serum SOD(P<0.05),increase the content of serum MDA(P<0.05),and increase the expression of UCP4 mRNA and protein in hypothalamus(P<0.05).Compared with the model group,hanjingtang could lower blood pressure(P<0.05),and further increase the level of plasma T3,T4,TSH,TRH(P < 0.05),increase the content of serum SOD,decrease the content of serum MDA,and furtherincrease the expression of UCP4 mRNA and protein in hypothalamus(P <0.05).Conclusions:1 Combined stimulation of cold and high salt diet can increase blood pressure in rats,and hanjingtang can reduce blood pressure.2 The hypobaric mechanism of hanjingtang may be related to the activation of HPT axis,which may promote the secretion of UCP4,increase heat production,and play a therapeutic role in treating hypertension with cold coagulation syndrome.3 Hanjingtang can increase the antioxidant capacity of the body,which may play a therapeutic role in this kind of hypertension.
