The Therapeutic Of Herbs Of Supplementing Qi And Nourishing Yin Stasis-removing And Collaterals-dredging And Its Disassembled On Dfl Rats And The Effect Of The Herbs On The Expression Of Liver Tissue SREBP2 And HMGCR

Objective:The effects of herbs of supplementing qi and nourishing yin stasis-removing and collaterals-dredging and its disassembled on Sterol regulatory element binding proteins 2(SREBP2)、 hydroxymethyl glutaryl coenzyme A reductase reducing ferment(HMGCR)and steatosis of DFL rats were observed,and the therapeutic effect and the possible mechanism were discussed.Methods:55 healthy male SD rats were randomly divided into control group(N group,n=8)and modelling group(n=47).Control group and modelling group were fed with normal diet and high fat and high carbohydrate diet separately.After 4 weeks,the model of diabetic rats was established with streptozotion(STZ)by intraperitoneal injection(35 mg/kg).After 72 h,the rats whose three times of random blood glucose(RBG)from tail vein were ≥16.7mmol/L were considered as successful model.The 41 rats were randomly divided into model group(M group),supplementing qi and nourishing yin resolving masses and collaterals-dredging group(ZYgroup),blood-promoting,stasis-removing and collaterals-dredging group(HT group),stasis-removing group(H group)and collaterals-dredging group(T group).The model group were 9 rats,and other groups were 8 rats.After given by intragastric administration for 16 weeks,the rats were killed.Serum TG and TC were determined by biochemical method.The morphological changes of liver tissues in rats were observed by HE staining.Real-time PCR and Western Blot were used to detect mRNA and protein expression of liver tissue SREBP2 and HMGCR.Results:HE staining indicates: In M group,different sizes of lipid droplets and ballooning change can be seen in the liver cell and the structure of hepatic lobule arranged in disorder.Compared with M group,the treatment groups’ degree of lipid degeneration was reduced.The content of TG and TC in the serum of rats: Cmpared with N group,M group and each trentment group increased obviously(P<0.01);Compared with M group,each trentment group reduced obviously(P<0.01);Comparison between Chinese medicine groups: ZY group is lower than HT、H、T groups obviously(P<0.05 or P<0.01);HT group is lower than H and T groups obviously(P<0.05 or P<0.01);there was no significant difference between group H and T group(P>0.05).The mRNA and protein levels of SREBP2 and HMGCR:Cmpared with N group,M group and each trentment group increased obviously(P < 0.05 or P < 0.01).Compared with M group,each trentment group reduced obviously(P<0.05 or P<0.01);Comparison between Chinese medicine groups: ZY group is lower than HT、H、T groups obviously(P<0.05 or P<0.01);HT group is lower than H and T groups obviously(P<0.05 or P< 0.01);there was no significant difference between group H and T group(P>0.05).Conclusion:The herbs of supplementing qi and nourishing yin stasis-removing and collaterals-dredging can improve the general status of DFL rats,and the possible mechanism is related to the expression of SREBP2 and HMGCR.The herbs of supplementing qi and nourishing yin stasis-removing and collaterals-dredging is superior to all the disassembled group.The herbs of stasis-removing and collaterals-dredging is superior to single stasis-removing group and collaterals-dredging group.The compatible application of herbs of supplementing qi and nourishing yin、stasis-removing and collaterals-dredging has synergistic effect.
