| Objective:This study aimed to observe the effect of chronic intermittent hypobaric hypoxia(CIHH)on iron metabolism in obese rats by establishing a high-fat diet and fructose-induced obesity model and to explore its mechanism.Methods:1 Experimental grouping: Adult male Sprague-Dawley rats were randomly assigned to four groups: obese group(DIO),CIHH group,CIHH-treatment obese group(DIO+CIHH)and control group(CON).Rats in CIHH and DIO+CIHH groups were placed in a hyperbaric chamber.However,rats in CON and DIO groups did not accept any treatment and were constantly placed in normoxic environment.2 The morphological changes of the liver,kidney and spleen were observed by HE staining.3 The levels of serum erythropoietin(EPO),ferritin(SF)and interleukin-6(IL-6)were detected by ELISA.4 serum iron(SI),total iron binding capacity(TIBC)and transferrin saturation(TS%)were assessed by spectrophotometry.5 Red blood cell(RBC)count and hemoglobin(Hb)content were detected by hematology analyzer Sysmex-xs500 i.6 The relative expression level of hepcidin mRNA in liver tissues was detected by RT-PCR.Results:1.Compared with CON rats,body weight of DIO rats was significantly higher(P<0.05),and no significant differences were noted in body weight in CIHH rats(P>0.05).Body weight of DIO+CIHH rats was significantly lower than that of DIO rats(P<0.05).2.Compared with CON rats,the results of HE staining in DIO rats showed that watery degeneration,fatty degeneration and liver cordiness in the liver,renal tubular epithelial cells bloated in the kidney,lymph nodules increased and lymphoid tissues proliferated significantly in the spleen;Pathology morphology in DIO+CIHH rats showed significant improved than that in DIO rats.3.Compared with CON rats,the contents of RBC and Hb in DIO rats decreased significantly(P<0.05),but there was no significant changes in CIHH rats(P>0.05).After CIHH treatment,RBC count and Hb content were significantly increased(P<0.05).4.Compared with CON rats,there was significantly increased TIBC and notably decreased SF in DIO rats(P<0.05),whereas there were no significant changes in serum TIBC,SI,TS% and SF in CIHH rats(P>0.05).SF was notably increased(P<0.05)in CIHH-treated DIO rats.5.Compared with CON rats,serum IL-6 level was increased significantly and serum EPO level was decreased in DIO rats(P<0.05),but IL-6 level and EPO level in CIHH rats did not differ significantly(P>0.05).IL-6 level was decreased remarkably and serum EPO level was elevated after CIHH treatment in DIO rats(P<0.05).6.Compared with CON rats,hepatic hepcidin mRNA expression in DIO rats were significantly increased(P<0.05),whereas that in CIHH rats did not differ significantly(P>0.05).Relative expression of hepcidin mRNA in DIO rats after CIHH treatment was remarkably decreased(P<0.05).Conclusion:CIHH plays a regulatory role in iron metabolism disorder in obese rat model,potentially by down-regulation of hepcidin via inhibiting IL-6 and promoting EPO expression. |
