| Objective:This topic through the study of angelica naphtha in spontaneously hypertensive rats(SHR)on the liver morphology,and the influence of lipid metabolism related genes in liver tissue Tnfaip8l2,Tlr3 expression,the influence of to discuss the effect of angelica naphtha in SHR lipid metabolism and its mechanism.Method:Take eight weeks of male SHR rats and the same weeks of wistar rats,before formal experimental pressure training once a day,7 d in a row,to adapt to the environment,when rats blood pressure stable,SHR were randomly divided into 3 groups,model group,atorvastatin calcium group and treatment group of angelica naphtha,Wistar rats as normal control group,each group 5 only.The model group and the normal control group were given normal saline and no medication.For atorvastatin calcium group and angelica volatile oil treatment group,the dosage was calculated according to the formula of rat body surface area,and the dosage was adjusted according to the change of body weight.The medicine is administered in the stomach for 4 weeks,7 days per week,and daily dose.After drawing blood,and killing rats,use HE dyeing hair and observe the liver tissue pathology slice change with inverted optical microscope and the changes of the ultrastructure of liver.The expression levels of Tnfaip8l2 and Tlr3 m RNA in rats were detected by q RT-PCR.The changes of expression levels of Tnfaip8l2 and Tlr3 protein in rats were detected by Western-blot.Data processing was analyzed using SPSS20.0 software.Results :1.Changes of blood pressure before and after medication: Before administration,The difference between the control group(P <0.05)was statistically significant.After four weeks of administration,there was no significant difference between the blood pressure of atorvastatin calcium group and the model group,and the difference was not statistically significant(P >0.05).After 4 weeks of continuous gavage,the blood pressure in the volatile oil group was significantly lower than that in the model group(P <0.05).2.Angelica naphtha effect on hypertension rats blood lipid levels: compared with model group,the treatment group of TC,TG,LDL-C level is lower than the model group,HDL-C is higher than that of model group,the treatment group after treatment,blood lipid level there are differences in change,but no significant difference.3.The effect of angelica volatile oil on the morphology of liver tissue in rats.Normal hepatic lobules in normal control group were normal.The model group showed fatty degeneration,and the liver cells had vesicles.The model group showed fatty degeneration,and the liver cells had vesicles.Compared with the model group,the glycogen decomposition and the disappearance of fatty degeneration were observed in the volatile oil group.There was no significant abnormality in atorvastatin calcium group.4,The effect of angelica volatile oil on the expression of m RNA and protein levels of Tnfaip8l2,Tlr3 gene in rats:Results show that compared with model group,Tnfaip8l2,Tlr3 the two genes in model group were not seen obvious abnormity,the expression of Tnfaip8l2 and Tlr3 is up-regulation in angelica group,atorvastatin calcium group,and that is diversity.The difference is statistically significant(P < 0.05).Conclusion:Angelica volatile oil reduces SHR’s blood pressure.Tnfaip8l2 and Tlr3 were up-regulated and differentiated in each drug group.Angelica volatile oil has regulatory effect on SHR fat metabolism,which may increase the m RNA level of Tnfaip8l2 and Tlr3 gene,inhibit the blood pressure of atherosclerosis,and affect lipid metabolism. |
