| Objective To explore the effects of PM2.5.5 emitted from motor vehicles on the proliferation function,apoptosis,DNA damage and DNA methylation in EA.hy926 cell line.Methods Motor vehicles PM2.5.5 was sampled using a fixed-source dilution channel and flow sampler.The PM2.5.5 suspension was extracted using an ultrasonic water-bath method and then human umbilical vein endothelial cell line(EA.hy926)were treated with various concentrations of it.Cell proliferation,apoptosis,DNA damage,and DNA methylation levels were measured for the cellular toxicity of PM2.5.Results The proliferation of EA.hy926 cell line is suppressed in 200μg/mL group and there was a statistically significant different(P<0.05)between control group and 200μg/mL group.The cell apoptosis rate and 8-OHdG%of EA.hy926 cell line was found increased and the5-mC%was found decreased.Conclusions PM2.5.5 emitted from motor vehicles can suppress viability,and cause 8-OHdG%increase and DNA methylation decrease in EA.hy926 cell line.There may no correlation between the variables. |
