Objective: The study is to evaluate the expressions of Yes-associated protein(YAP1)and Serine/arginine-rich splicing factor 3(SRSF3)in the tissue of cervical squamous cell carcinoma and investigate their association with the clinicopathological features of cervical carcinoma.And to explore the prognostic values of YAP1 and SRSF3 expression in cervical carcinoma.Methods: By using the Immunohistochemistry staining method,the expressions of YAP1 and SRSF3 in cervical squamous cell carcinoma and normal cervical tissue were detected.Spearman’s rank correlation,Kaplan-Meier plots and Cox proportional hazards regression model were used to analyze the data.Results:(1)The expression level of YAP1 and SRSF3 in cervical squamous cell carcinoma tissues was higher than that in normal cervical tissues(P <0.01).(2)The expression of SRSF3 was observed in the nucleus of koilocytes of paracancerous tissues and normal cervical tissues.(3)The expression of YAP1 and SRSF3 was correlated with lymphatic metastasis(P <0.05),but not with age,FIGO stage,histological differentiation,depth of interstitial infiltration,tumor size,margins and adjuvant therapy(P> 0.05).(4)There was a strong correlation between YAP1 protein expression in the cytoplasm and the nucleus(r = 0.800,P <0.01).There was a moderate correlation between YAP1 expression in the cytoplasm and SRSF3 protein expression(r = 0.696,P <0.01).There was a moderate correlation between YAP1 protein expression in the nucleus and SRSF3 protein expression(r = 0.662,P <0.01).(5)Univariate survival analysis showed that OS and DFS were affected by lymph node metastasis,margins,YAP1 and SRSF3(P <0.05),but not by patient age,FIGO stage,depth of interstitial infiltration,tumor size,histological grade and assisted treatment(P> 0.05).(6)Multivariate analysis showed that lymph node metastasis was an independent prognostic factor affecting overall survival(P = 0.042,RR = 3.985).In disease-free survival analysis,positive margins were independent adverse prognostic indicators(P = 0.017,RR = 6.620).Conclusion: YAP1 and SRSF3 were highly expressed in cervical squamous cell carcinoma,and the expression of YAP1 and SRSF3 was positively correlated.The expression of YAP1 and SRSF3 may be related to lymph node metastasis.The combined detection of these two proteins may provide some guidance for the clinical diagnosis and prognosis of persistent cervical cancer with HPV infection and provide clues for the development of new therapeutic approaches.YAP1 and SRSF3 proteins may be potential drug targets.
