| Objective: to investigate the expression of MCM3 gene in bladder cancer tissues,adjacent normal bladder tissues and bladder cancer cell lines,and to analyze the difference of MCM3 gene expression between normal and cancerous tissues.To elucidate the correlation between the expression level and the clinicopathological features of bladder cancer patients,and to confirm the role of MCM3 in the proliferation and invasion of bladder cancer cells through in vitro cell experiments.Methods: the expression of MCM3 m RNA in bladder cancer tissues and adjacent normal bladder tissues and bladder cancer cell lines 5637 and T24 were detected by real-time quantitative PCR.Meanwhile,real-time quantitative PCR and Western Blot were used to detect the expression of m RNA and protein in bladder cancer cell lines 5637 and T24.MCM3 specific si RNA was transfected into bladder cancer cell lines by liposome transfection.The role of MCM3 in the proliferation and invasion of bladder cancer cells was studied by CCK8 / Transwell assay.Results: the expression of MCM3 in clinical specimens was significantly higher than that in adjacent normal tissues,and was positively correlated with invasion,clinical stage,recurrence and metastasis of bladder cancer.At the cellular level,the expression of MCM3 increased with the increase of malignant degree of bladder cancer cells.The specific si RNA was transfected into the cells by RNA and protein detection,which confirmed that MCM3 was effectively knocked out.CCK8 assay showed that the proliferation of MCM3 knockout group was significantly inhibited compared with the control group.The results of Transwell assay showed that the invasion ability of MCM3 cells was significantly lower than that of the control group.At the same time,Western blot was used to detect the expression of proliferative and invasive-associated proteins in bladder cancer cells after MCM3 knockout.Conclusion as a special factor of DNA replication,the expression level of MCM3 can directly reflect cell proliferation,which is useful for the diagnosis and prognosis of various diseases such as atypical hyperplasia,proliferative lesion and tumor caused by cell cycle disorder and runaway.Of great significance.This experiment shows that MCM3 can promote T 24.5637 Bladder cancer cell proliferation and promote bladder cancer cell invasion,all these indicate that MCM3 plays the role of oncogene in bladder cancer cells,is a kind of oncogene of bladder cancer.Combined with the recent domestic and foreign studies on the progress of MCM3 in tumor progression and differentiation between benign and malignant tumors,it is basically confirmed that MCM3 plays an important role in the biological behavior of bladder cancer. |
