Objective: To explore treatment effect of human umbilical cord mesenchymal stem cells(HUC-MSCs)transplantation in different ways on acute liver failure(ALF)in rats.Methods: 60 SPF grade SD rats were randomly into 4 groups according to the principle of complete randomization: caudal vein transplantation group,hepatic lobe injection transplantation group,model group and control group.HUC-MSCs Detected by Cell Morphology,Cell Markers and Cell Differentiation.HUC-MSCs were isolated and identified by flow cytometry.Number of HUC-MSCs transplantation in treatment group was 1.0×106.Liver function test:Detection of ALT,TBil,and ALB levels at 0,1,2,4 and 7 days after cell transplantation.Histopathological observation: Liver specimens were prepared at 3 days,1 week and 2 weeks after injection of HUC-MSCs to observe the pathological changes of liver cells.Detection of human CK18 expression by immunohistochemistry to observe its colonization in the liver.Real-time PCR assay was performed to detect transcriptional levels of CK8,CK18 and AFP genes.Results:1.Three days after transplantation,serum levels of TBil and ALT in the liver transplantation group were significantly lower than those in the model control group,but ALB was the opposite(P < 0.05).2.HUC-MSCs improved histological status through enhancing hepatocellular regeneration and reducing inflammatory cells In the treatment group.It shows that HUC-MSCs have a repairing effect on the liver of ALF rats.3.Three days after transplantation,the expression of human CK18 protein was detected in the treatment group.It indicates that HUC-MSCs can survive in rat liver tissue and further be directed to differentiate into cells with the ability to secrete proteins and surface specific markers.4.A week after modeling,the hepatic function and differentiation of stem cells in the caudal vein transplantation group and the hepatic lobe injection transplantation group were similar,and the difference was not statistically significant(P> 0.05).Conclusion:1.Transplantation of HUC-MSCs can improve liver function and promote liver repair after ALF and play a therapeutic role.2.Transplantation of HUC-MSCs through the tail vein has similar therapeutic effects to intrahepatic injection,can repair liver tissue.3.HUC-MSCs can survive in rat liver tissue and further be directed to differentiate into cells with the ability to secrete proteins and surface specific markers.
