Objective:To explore the functions of different concentrations of IL-6 on the expression of Janus kinase 1(JAK1),Signal Transducer and Activator of Transcription 3(STAT3)and Cyclin D1 in prostate cancer cell lines PC-3 and DU 145.According to the results,we discuss the activation of the key target protein on the signal pathway and the regulation of the downstream target protein on the signal pathway,and some new mechanisms in the process of the development of prostate cancer are described.Methods:1.After treating prostate cancer cell lines PC-3 and DU 145 by IL-6 in different concentrations,we detected the cells proliferation with CCK-8.2.After treating prostate cancer cell line PC-3 and DU145 by IL-6 in different concentrations,we detected the expression of Cyclin D1 in two cell lines with real-time fluorescent quantitative PCR.3.After treating prostate cancer cell lines PC-3 and DU 145 by IL-6 in different concentrations,we detected the expressions of JAK1,STAT3 and Cyclin D1 in two cell lines with western bolt.Results:1.The effects of IL-6 on proliferation of prostate cancer cells PC-3 and DU 145(1)The CCK-8 method was used to measure the proliferation on PC-3 cell lines with the different IL-6 concentrations of 0ng/ml,50ng/ml and 100ng/ml.In 12hs and 24hs,there were significant differences in the OD values between the experimental groups 50ng/ml,100ng/ml and the control group Ong/ml.The results showed that IL-6 could promote the proliferation of PC-3 cell lines.(2)The CCK-8 method was used to measure the proliferation on DU145 cell lines with the different IL-6 concentrations of Ong/ml,50ng/ml and 100ng/ml.In 12hs and 24hs,there were significant differences in the OD values between the experimental groups 50ng/ml,100ng/ml and the control group Ong/ml.The results showed that IL-6 could promote the proliferation of DU 145 cell lines.2.The effects of IL-6 on the expression of Cyclin D1 in PC-3 and DU 145 cell lines on transcriptional levelThe results showed that the expression of Cyclin D1 was increased on transcriptional level in PC-3 and DU 145 cell lines after treating with different concentrations of IL-6 for 24hs.The more expression of Cyclin D1 correlated with the higher concentration of IL-6.3.The effects of IL-6 on the expressions of JAK1,STAT3 and Cyclin D1 in PC-3 and DU 145 cell lines on translational level(1)The effects of IL-6 on translation level expression of JAK1 in PC cell lines PC-3 and DU145:We found that the translational expression of p-JAK1 in PC cell lines after treating with different concentrations of IL-6 for 24hs was different.The more expression of p-JAK1 correlated with the higher concentration of IL-6.(2)The effects of IL-6 on translation level expression of STAT3 in PC cell lines PC-3 and DU 145:The results were that the translational expression of p-STAT3 in PC cell lines after treating with different concentrations of IL-6 for 24hs was different.The more expression of p-STA-T3 correlated with the higher concentration of IL-6.(3)The effects of IL-6 on translation level expression of Cyclin D1 in PC cell lines PC-3 and DU 145:The study expored that the translational expression of Cyclin D1 in PC cell line after treating with different concentrations of IL-6 for 24hs was different.The more expression of Cyclin D1 correlated with the higher concentration of IL-6.Conclusions:1.IL-6 promotes the proliferation of androgen-resistant prostate cancer cell lines PC-3 and DU145.2.The transcriptional expression of Cyclin D1 in PC-3 and DU145 lines is significant after treating with different concentrations of IL-6 for 24 hs.The expression of Cyclin D1 correlates with the higher concentration of IL-6.3.The translational expressions of p-JAK1,p-STAT3 and Cyclin D1 in PC-3 and DU 145 cell lines are significant after treating with different concentrations of IL-6 for 24hs.The expressions of p-JAK1,p-STAT3 and Cyclin D1 correlate with the higher concentration of IL-6.4.1n the process of IL-6 promoting the proliferation of prostate cancer cells,JAKl/STAT3/CyclinDl may be an important regulatory pathway involved.
