[Objective] To observe the expression of MBD2,SOCS3 and STAT3 in the lung tissue and spleen cells of severe asthmatic mice and to explore the role of MBD2 and SOCS3 in the pathogenesis of severe asthmatic mice.[Methods] 1.30 mice were randomly divided into 3 groups,10 rats in each group,The severe asthmatic mice model was established with 100 μg HDM + 100 μg OVA + 15 μg LPS,the conventional asthma mouse model was established with 25 μg OVA,and the control mice were established by normal saline.The mice’s lung function,BALF cell count and lung histopathology were detected.The concentrations of IL-17 in serum and BALF in each group were detected by ELISA.(2)The expression of MBD2,SOCS3 and STAT3 in lung and spleen cells of mice were detected by Western-blot Methods;3.The expression of MBD2,SOCS3 and STAT3 in lung and spleen cells of three groups were detected by PCR.[Results] 1.Compared with the control group and the conventional asthma group,the mice in the severe asthma group had lethargy.Airway resistance was significantly increased in severe asthmatic mice when stimulated by methacholine,the total number of the cells and the number of neutrophils in BALF were the highest,and the inflammatory reaction in lung tissue was the most obvious.The highest level of IL-17 was found in the serum and alveolar lavage fluid of severe asthmatic mice.2.Western-blot results showed that compared with the control group,the expression of MBD2 and STAT3 protein in the splenocytes of the conventional asthma group and the severe asthma group increased,especially in severe asthma group(P<0.05).Compared with the control group,there was no significant difference in the expression of SOCS3 protein in the spleen cells of mice with severe asthma.Compared with the conventional asthma group,the expression of SOCS3 protein in the spleen cells of the severe asthmatic mice was decreased.3.The result of Western-blot showed that the expression of MBD2 and STAT3 in lung tissue of conventional asthma group and severe asthma group increased compared with the control group,and the highest in severe asthma group(P<0.05).The expression of SOCS3 in lung tissue of severe asthma group was lower than that in control group and conventional asthma group(P<0.05).4.Spleen cell PCR showed that the relative expression of MBD2 gene in conventional asthma group and severe asthma group was higher than that in control group,while it was the most obvious in severe asthma group(P<0.05).Compared with the control group,STAT3 and SOCS3 expression in conventional asthma group and severe asthma group had no significant difference(P>0.05).5.Lung tissue PCR showed that the MBD2 in severe asthma group was higher than that in control group and conventional asthma group,while the relative expression of SOCS3 was lower than that in control group and conventional asthma group(P<0.05).There was no significant difference in the relative expression of STAT3 gene between the three groups of mice(P>0.05).[Conclusion] 1.The expression of MBD2 in lung tissue and spleen cells of severe asthmatic mice increased,while that of SOCS3 was low.2.The involvement of MBD2 and SOCS3 in the pathogenesis of severe asthma may be related to the regulation of Th17 cell differentiation.
