BackgroundMissed abortion as known as expired abortion,is a special case of natural abortion.It refers to the failure of the embryo or fetus to be expelled in time.It can be manifested as the disappearance of the early pregnancy reaction,often symptoms of threatened abortion,or without any symptoms.The uterus no longer increases with the increase of gestational age.The cause of missed abortion is complicated and unknown,and induce spontaneous abortion are also involved in embryo factors,maternal factors,immune dysfunction and environmental factors.According to the survey,chromosomal abnormalities are one of the most common causes of early abortion,more than half of which are related to chromosomal abnormalities.Study on missed abortion embryo chromosome,clear for missed abortion has a guiding role in good condition,at the same time,the analysis of chromosomal abnormalities,can be better for the next time to prepare for a healthy pregnancy.Array comparative genomic hybridization(array-CGH)is an efficient novel molecular genetic analysis method developed,its advantage lies in the cultivation of cells without direct extraction of genomic DNA samples,were detected by a hybridizationexperiment to genome-wide DNA copy number variants,(CNVs)were analyzed with high resolution,high and accurate detection of chromosomal imbalances in the submicroscopic structure of traditional karyotype analysis that can not be detected by,and can be a variation of DNA sequences accurately locate on chromosome,the chromosomal disease to gene level.It has the advantages of high resolution,short detection time,high degree of automation,by comparing with various disease database,to determine the pathogenicity of CNVs,provides a new direction for disease of unknown cause of missed abortion genetic diagnosis,provide an effective molecular genetic diagnosis method.This paper mainly through the 72 cases of missed abortion were fetal chromosome analysis,explore the relationship between chromosomal abnormalities and missed abortion.Objective1.To analyze the proportion of abnormal chromosomal abnormalities in missed abortion patients.2.Analyze the proportion of chromosomal abnormality in different types of embryos,3.To explore the application value of chromosome microarray analysis in detecting the genetic variation of missed abortion.Methods1.The embryo chorionic chromosomes were collected from the patients with aborted abortion.2.Analysis the missed abortion fetal chromosome abnormalities with the chromosome gene chip analysis(CMA)3.The results obtained by statistical analysis are discussed.Results1.In 72 cases of fetal villus chromosome analysis,the villus specimens were all detected successfully,34 cases were normal,accounting for about 47%of the total specimens,38 cases were abnormal,and the abnormal detection rate was 53%.(table 3-2).2.Among the samples of chromosomal abnormalities,32 were chromosomal abnormalities,accounting for 84.2%of the abnormal number of chromosomes,4 were chromosomal abnormalities and accounted for 10,5%of the abnormal number of chromosomes,2 cases were chimerism,accounting for 5.3%of abnormal chromosomes,(table 3-3).3.Compared with the traditional karyotype analysis,the chromosome microarray analysis technique has a higher success rate,shorter experimental period and higher resolution,which can provide more genetic variation information than the traditional karyotype analysis.Conclusion1.Chromosomal abnormality is one of the main causes of missed abortion.2.The most common type of chromosomal abnormality is chromosomal abnormality,followed by chromosomal structural abnormalities,the last is chimeric body.3.Compared with the traditional karyotype analysis,the technique of chromosome microarray analysis has the advantages of higher detection success rate,shorter experimental period and higher resolution.
