Objective: To collect and classify the intestinal flora of the Choledocholithiasis before and after ERCP,the relationship between intestinal microflora and the formation of cholelithiasis was investigated,and the effect of ERCP operation on the intestinal flora of the patients was also understood.Methods: Collect the fecal specimen who were hospitalized in our hospital from December 2016 to October 2017,to collect their feces.The bacterial genome DNA was extracted from the fecal bacteria,and the primers were designed according to the 16 SrDNA gene sequence of the bacteria.The changes of bacteria were detected by the RT-PCR method.Furthermore,we compared the intestinal microflora genome of 38 normal people to clarify whether there were differences in the composition of intestinal flora between normal and gallstone patients.Results: In this study,we obtained a total of 157,721 high-quality,categorizable bacteria 16 S rRNA gene sequences from all samples and identified 4652 bacterial OUT numbers.Finally,11 bacterial taxa were identified from patients with gallstones,of which Firmieutes,Proteobacteria,and Bacteroidetes accounted for at least 75% of the constituent ratios,and120 bacteria.Genera,of which Enterococcus,Escherichia/Shigella,Bifidobacterium,Bacteroides,Faecalibacterium accounted for the composition More than 30%.It was found that in the level of bacteria,the intestinal Proteobacteria in patients with gallstones was significantly higher than normal,at the level of bacteria Three species of Roseburia,Lachnospiracea,and Faecalibacterium were significantly different from normal individuals in gallstone patients(P<0.05),which was significantly lower than that of normal people.However,there was no significant difference in intestinal flora between the two groups before and after ERCP,P>0.05.Conclusion: The diversity of human intestinal flora,intestinal flora imbalance and cholelithiasis formation have certain correlation.The intestinal microflora in patients with choledocholithiasis has no obvious change after ERCP.
