Objective:Through the analysis of the correlation between polymorphism of ABCG2 gene and genotype of ABCG2 gene in gout and hyperuricemia population in southwestern Hubei Province,we further provided basis for genetic and polymorphism differences in gout and hyperuricemia populations.The research and development of new gout drugs in various regions provide reference.Methods:Gene sequencing and biochemical indexes were collected from 158 cases of gout,189 cases of high uric acid and 306 cases of normal group.The rs2231142 fragments amplified by PCR were analyzed by high resolution dissolution curve(HRM)method,and the single nucleotide polymorphisms were detected.The genotype was verified by gene sequencing and statistical analysis was made.Results:Accurate and reliable.Among 158 cases of gout group,48 cases were AA type,accounting for 30.30%;189 cases of high uric acid group included 22 cases of AA type,accounting for 11.64%;306 cases of normal group,including 28 cases of AA type,accounting for 9.15%;AA genotype carrier in normal group The rate was significantly lower than that of gout group.The higher the carrying rate of AA genotype,the higher the risk of gout,which was increased by 7.584 times(p<0.05).The frequency of genotype A in the gout group accounted for 54.75%,which was 20.36%higher than that of the normal group(x~2=48.833a,p<0.05).The frequency of alleles in the hyperuricemia group A accounted for 34.39%,which was 3.67%higher than that of the normal group.(x~2=1.777a,p=0.411).It can be seen that the frequency of A alleles is positively correlated with the incidence of gout,and the risk of gout is significantly higher than that of the C allele.The genes in the hyperuricemia group are compared with the normal group.There was no significant difference in the frequencies of type and alleles carried.The values of biochemical indexes in the normal group were significantly lower than those in the gout group and the hype ruricemia group(p<0.05).Conclusion:The gene polymorphism of rs2231142 in ABCG2 gene was associated with gout in the population in southwestern Hubei;There were regional differences in genotype and allele carrying rates,which provided an important genetic marker for the early diagnosis of gout in the population of Southwest Hubei Province.Gout correlated with related biochemical indicators,further confirming that gout and hyperuricemia increased the risk of diseases such as diabetes,heart disease,and liver and kidney damage compared with the normal group,and their mutual influence and concomitant relationship;established A rapid and economical PCR-HRM technique is used to detect polymorphisms in the ABCG2 gene of gout and hyperuricemia,and it is also beneficial for large-scale molecular epidemiological studies of ABCG2 gene-related diseases.
