| ObjectiveThe study built the model of venous indwelling needle in animal,intervened in the peripheral vascular of indwelling needle through the external Anisodamine,tested the marker associated with blood coagulation function D-Dimer(D-D),the marker associated with vascular endothelial cell injury Vascular endothelial cell growth factor(VEGF),Inflammatory factor tumor necrosis factor(TNF-a),the makers associated with blood rheology activated partial thromboplastin time(APTT),prothrombin time(PT),fibrinogen(FIB),thrombin time(TT),the platelet count(PLT),mean platelet volume(MPV),platelet volume distribution width(PDW),platelet hematocrit(PCT)level at the time before catheter and after catheter.Analysis by combining with the naked eye of phlebitis and the pathological results,to determine the situation of Vascular endothelial injury and the incidence of thrombosis by indwelling needle.To reveal the effect of anisodamine intervention in venous indwelling needle.And to interpret the mechanism of anisodamine intervention in thrombosis.Method1.The rabbits were randomly assigned to blank group(n= 10,)the physiological sal:ine group(NS group,n=30),the saline Anisodamine group(NA group,n=30),Mannitol group(M group,n=30)and Mannitol Anisodamine group(MA group,n=30),NS group,NA group,M group,MA group were intravenous indwelling needle,in accordance with the indwelling time is divided into 1D,3D,5D of 12 groups.2.In group NS,group NA infusion of 0.9%saline solution,two times a day.In group M,group MA infusion of 20%Mannitol solution,two times a day.10 minutes before infusion Anisodamine ointment were rubbed into vessel surface.3.To observe the phlebitis after indwelling needle.blank group,the physiological saline group,the saline Anisodamine group,Mannitol group and Mannitol Anisodamine group were tested the blood D-D,VEGF,TNF-a,APTT,PT,FIB,TT,PLT,MPV,TNF-a,PDW,PCT value at the different time,which time before catheter and after catheter 1 day,3 day,5 day.Each group ribbits were killed according to the retention time,to make ear vein tissue sections,and to observe the form and pathological changes of venous thrombosis under the light microscope.Result1.Catheter for 3 days,serum D-D concentration,D-D concentration decreased in group MA.NS group TNF-a concentrations increased after catheter 3 days in group NA decreased,TNF-aconcentration decreased than before catheter,catheter for 5 days increased the TNF-aconcentration in M group,MA group,TNF-aconcentration decreased.2.Catheter for 1 days,1 days of catheter was statistically significant blank group and NA group,MA group,VEGF(P<0.05)concentration difference.Catheter for 5 days,VEGF were Almost equal,almost recovered to levels before catheter.3.Catheter for 5 days,the blank group and the four group comparison of APTT catheter group were significantly different,the difference was statistically significant.Groups PT,FIB,TT,PLT,MPV,PDW,PCT levels in the range of reference values,the difference was not statistically significant.4.Catheter for 3 days,indwelling catheter phlebitis was 100%,M group’Ⅱ level was hinger than MA group of phlebitis in the two groups have statistical significance level of difference(P<0.05).5.The middle section of the most severe needle thrombosis,catheter thrombosis increased duration.6.Thrombosis related positively with the content of FIB,the relationship between the concentration of D-D was positively correlated with APTT,there was a negative correlation between TNF-a and PLT.Conclusion1.Anisodamine interfered had changed the concentration of TNF-alpha,D-D,VEGF,thus affected the formation of thrombus,VEGF appeared earlier changed suggestive of thrombosis.2.Effects of Anisodamine on PT,APTT,TT,FIB and PLT,MPV,PDW,PCT,coagulation related indicators of intervention effect was not obvious.3.Thrombosis cauded by needle can not accurately judge by visual indwelling venous,actually appeared in the local reaction vessels and surrounding tissue,the pathological damage has occurred.4.Incidence of phlebitis by venous indwelling needle had increased with the prolonged retention time,the more serious the degree was increased,and the severity of venous thrombosis.5.Catheter induced thrombosis and D-D,TNF-alpha,VEGF,PT,APTT,TT,FIB,PLT,MPV,PDW,PCT index had certain correlation between. |
