| Objective: To explore the effects of biological characteristics index of the anogenital distance(AGD),body weight(BW),main organ weight and its organ coefficients,serum sex hormones and estrogen receptor,male testis development after birth involve in Kunming mice(KM Mice)during pregnancy of Bisphenol A(BPA)exposure.Methods: We selected thirty healthy female KM mice,which were randomly divided into six groups: negative control group(corn oil group),positive control group(diethylstilbestrol group),low dose group of Bisphenol A(2 mg/kg/d),middle dose group of Bisphenol A(20 mg/kg/d),high dose group of Bisphenol A(200 mg/kg/d)and Bisphenol A 20 mg/kg/d combine with soy isoflavones 300 mg/kg/d group.From pregnancy(GD)9 to the day before the offspring birth,the pregnant mothers were treated by group gavage with intragastric administration.To determine postnatal day(PND0),PND7,PND14,PND21 and PND26 AGD of the offspring;PND0,PND14 and PND26 BW of the offspring;the organ weight(thymus,liver,spleen,testis or uterus);detecte the content of sex hormone and estrogen receptor in serum of PND26 offspring mice with ELISA;perform PND26 testicular histopathological section and observe its pathological morphology.Results: In the offspring,compared with the control group,there was no significant change in the number and sex ratio with the other dose groups;from PND0 to PND26,the BW growth and AGD increase in the offspring,compared with the control group,bisphenol A low-medium-high dose group and bisphenol A 20 + soy isoflavone 300 group increased the weight and AGD distance of the offspring significantly(P <0.05);compared with the control group,the medium and high dose of bisphenol A decreased the content of estrogen receptor(P <0.05);compared with the control group,the weight of the offspring in the low-dose BPA group increased in PND26,and the liver weight and liver coefficient increased significantly(P <0.05);compared with the control group,the high dose of bisphenol A decreased the content of estrogen receptor(P <0.05);Bisphenol A 20 + soy isoflavone 300 group increased the weight of the thymus and spleen,as well as its organ coefficient and decreased the content of estrogen receptor and T of the offspring(P <0.05);PND26 testicular pathology revealed that low,medium and high doses of BPA caused earlier proliferation and differentiation of germ cells.Conclusions: The endoestrogen effect produced by bisphenol A exposure can lead to the offspring mice abnormal growth and development of the body weight,the length of anogenital distance and liver,as well as decrease the estrogen receptor content and early proliferation and differentiation of testicular germ cells. |
