Objective:To study the inner particle formation cells through the miR-330/CNTTB1/CyclinD1 pathway to regulate cutin.Methods:1.The difference of mRNA expression of CTNNB1 between psoriatic patients and healthy people by qPCR.2.To determine the mRNA and protein levels of CTNNB1 in HaCaT cells in IL-22 and in HKC.Western blotting was used to determine the response to the inhibition of IL-22 and miR-330 in co-treatment.The wild-type and mutant CTNNB1mut-CTNNB1)luciferase reporter gene vector with 5 BP mutation at the predicted miR-330 binding site was constructed by searching for miRBase4.5.Double luciferase assay was used to determine whether miR-330 and CTNNB1 could be targeted binding.LV-miR-330 was used to infect HaCaT cells and HKCs,and protein levels of cyclin D1 and Axin2 in HaCaT cells and HKC were determined by Western blotting.Detection of cyclin D1 and Axin2 in HaCaT cells and HKC was performed.Protein and cell level in miR-330/CNTTB1/CyclinD1 pathway mediated by Zhuhuang granule.Conclusion:1.The expression of CTNNB1 mRNA in psoriatic lesions showed a downward trend.2.IL-22 increased the expression of mRNA and protein of CTNNB1 in keratinocytes.MiR-330 could inhibit the IL-22 induced increase in CTNNB1 protein.3.MiR-330 could directly target and regulate CTNNB1 expression 4.miR-330/ CTNNB1 / CyclinD1 pathway.Zhuhuang granules can regulate the formation of keratinocytes by miR-330/CTNNB1/CyclinD1 pathway.A cypress.
