ObjectiveTo detect the expression difference of lncRNA GAS-5 and TUG-1 in peripheral blood lymphocytes of RA active patients and normal persons,and to explore the correlation between lncRNA GAS-5 and TUG-1 and RA immune function and inflammatory markers,and to explore its clinical significance.MethodCollected physical examination patients in rheumatology and health medicine department from The hospital of Hubei University for Nationalities,from January2017 to November January 2017,and detected the data of RF,CRP and ESR by automatic biochemistry analyzer.By using reverse transcriptase ds-cDNA synthesis kit provided by Invitrogen company,synthetic double stranded complementary DNA;with GAPDH as reference,the expression difference of relative quantitative method for Real-time PCR lncRNA GAS-5 verification and TUG-1 into the SYBR Green fluorescent dye doped.Statistical software SPSSl7.0 was used.Mean + standard deviation measurement data(x±s)form,using t test;analysis of the differences between groups by ANOVA,P<0.05 had significant difference,P<0.01 had significant difference.Results1.The expression level of Lnc RNA GAS5 in peripheral blood lymphocytes of patients with rheumatoid arthritis was 0.994,while that of healthy controls was 2.901;the expression of LncRNA GAS5 in peripheral blood lymphocytes of RA patients was compared with that of healthy controls.Significantly lower(P<0.000);2.The average expression level of LncRNA TUG1 in peripheral blood lymphocytes of patients with rheumatoid arthritis was 0.135,while that of healthy controls was 0.673;the expression level of LncRNA TUG1 in peripheral blood lymphocytes of RA patients and healthy controls.The group was significantly lower(P < 0.000);3.There was no correlation between ESR,C-reactive protein levels and LncRNAGAS5 and TUG1 expression levels;4.Rheumatoid factor was negatively correlated with LncRNA GAS5 and TUG1(r=-0.699,P=0.000<0.05;r=-0.513,P=0.029<0.05);ConclusionIn the development of rheumatoid arthritis,LncRNA GAS5 and TUG1 are likely to play an extremely important role in providing strategic support for the effective diagnosis and treatment of RA;further study of lncRNA.It provides important reference for the molecular mechanism of RA pathogenesis,discovery of new diagnostic markers and therapeutic targets,and has important theoretical research value and clinical application prospects.
