| Objective: To investigate the association of the two functional polymorphisms in matrix metalloproteinase(MMP)genes(MMP-10 rs17435959(C>G),MMP-13 rs2252070(G>A))with the risk of chronic obstructive pulmonary disease(COPD)in a Chinese population.Methods: 305 cases and 326 controls obtained matching on Age,gender and smoking status and were successfully genotyped.The genotypes of the two polymorphisms were determined by polymerase chain reaction(PCR)based on the method of base-quenched probe.Results: For the rs17435959(C>G)of MMP-10 gene,no significant differences were found in allele(P=0.66,OR=0.90)and genotype frequencies(P=0.74,OR=1.08)between COPD patients and controls,nor was in the subgroup analysis(mild and moderate COPD vs.severe and very serve COPD vs.healthy control,allele frequency P=0.89,genotype frequency P=0.83).For the rs2252070(G>A)of MMP-13 gene,no significant differences were found in allele(P=0.76,OR=0.96)and genotype frequencies(P=0.58,OR=0.90)between COPD patients and controls,nor was in the subgroup analysis(mild and moderate COPD vs.severe and very serve COPD vs.healthy control,allele frequency P=0.94,genotype frequency P=0.36).The test of association between the four haplotypes and COPD was not significant,however an increased frequency of the G-G haplotype in MMP-10 and MMP-13 gene was observed in serve and very serve COPD patients compared to healthy controls(OR = 1.86;P=0.099).Conclusion MMP-10 rs17435959(C>G),MMP-13 rs2252070(G>A)in the Chinese population were polymorphic,however this two polymorphisms were lack of association with the risk of chronic obstructive pulmonary disease in the Chinese population.The G-G haplotype of MMP-10 rs17435959(C>G)and MMP-13 rs2252070(G>A)may increase the risk of serve and very serve COPD and which indicated a need for further study. |
