Study On The Relationship Between The Content Of Mogroside V And The Relieving-Cough,Expectorant And Laxative Efficacy

Objective: To investigate the correlation between the content of Mogroside V,the color and taste of fruit pulp,the origin,fruit growth period,medicinal product specifications and the efficacy of Siraitia grosvenorii relieving-cough,expectorant and laxative.The effects of pharmacological of the content of Mogroside V on cough,sputum and constipation,clearly define the main factors that affect the efficacy of Siraitia grosvenori and improve the quality evaluation system of Siraitia grosvenori.Methods:1.Pharmacological effects of extracts of Siraitia grosvenori water extraction and alcohol precipitation on cough,sputum and constipation: The relieving-cough effect of 37 batches of the extracts of the water extraction and alcohol precipitation of Siraitia grosvenori was determined by the coughing method of ammonia water;the expectorant effect of 37 batches of the extracts of the water extraction and alcohol precipitation of Siraitia grosvenori was determined by the mouse tracheal phenol red method;the laxative effect of 37 batches of the extracts of the water extraction and alcohol precipitation of Siraitia grosvenori was determined by the compound difenoxate-induced constipation model in mice.Statistical analysis of Siraitia grosvenori water extraction and alcohol precipitation has relieving-cough,expectorant and laxative efficacy.2.Determination of Mogroside V content: Using High Performance Liquid Chromatography,the content of Mogroside V in 37 batches of Siraitia grosvenorii(dried fruit)was determined.3.Determination of Siraitia grosvenorii color and taste of fruit pulp: The color and taste in 37 batches of Siraitia grosvenorii(dried fruit)fruit pulp were determined by colorimetry and mouse taste.4.According to results of determination of the content of Mogroside V,the color,taste,origin,growth period of fruits and the medicinal product specifications in 37 batches of Siraitia grosvenorii(dried fruits),statistical analysis of the correlation between the content of Mogroside V etc,and the efficacy of relieving-cough,expectorant,laxative.Results:1.The pharmacological effects of relieving-cough,expectorant and laxative of water extraction and alcohol precipitation from Siraitia grosvenorii: In 37 batches of extracts of Siraitia grosvenorii water extraction and alcohol precipitation,the effective rate of cough relieving was 72.97%,the effective rate of sputum was 70.27%,and the effective rate of constipation was 89.18%.2.Determination of Mogroside V content: 37 batches of Siraitia grosvenori(dry fruit),Mogroside V content equal to or greater than 0.50% of 35 batches,Mogroside V content less than 0.50% there are 2 batches.3.Determination of the color and taste of Siraitia grosvenorii fruit pulp: 37 batches of Siraitia grosvenorii fruit pulp color,34 light-colored batches and 3 deep-colored batches;37 batches of Siraitia grosvenorii fruit pulp,there are 21 sweet batches,16 non-sweet batches.4.Analysis on the correlation between Mogroside V,the color and taste of fruit pulp and the relieving-cough,,expectorant and laxative efficacy:(1)The correlation between Mogroside V content and the efficacy of cough,sputum,and laxative: The Mogroside V content is greater than 0.50% in the group and the cough effective rate is 77.14%;the content is less than 0.50% in the group and the cough effective rate is 0%.In the two groups.the difference was significant(P<0.05);the Mogroside V content is greater than 0.50% in the group and the expectorant effective rate is 62.86%;the content is less than 0.50% in the group and the expectorant effective rate is 100%.There was no significant difference between the two groups(P>0.05).);the Mogroside V content is greater than 0.50% in the group and the laxative effective rate is 88.57%;the content is less than 0.50% in the group and the laxative effective rate is 100%.There was no significant difference between the two groups(P>0.05).).(2)The correlation between the color of Siraitia grosvenorii fruit pulp and the efficacy of relieving-cough,expectorant and laxative: The effective rate of relieving-cough was 70.59% in light-colored group and 100.00% in dark-colored group.There was no significant difference between the two groups in cough efficiency(P>0.05);light-colored expectorant effective rate 67.65%,dark-color expectorant effective rate 66.67%,the two groups expectorant effective rate,the difference was not significant(P> 0.05);light-colored group constipation effective rate was 91.18%.The efficiency of constipation was 66.67% in the dark-colored group.There was no significant difference between the two groups(P>0.05).(3)The correlation between the taste of Siraitia grosvenorii fruit pulp and the efficacy of relieving-cough,expectorant and laxative: The effective rate of relieving-cough was 66.67% in the sweet group and 81.25% in the non-sweet group,and the relieving-cough efficiency was comparable in the two groups.Significant(P>0.05);Effective rate of 61.90% for sweet group and 75.00% for non-sweet group,no significant difference between two groups(P>0.05);Sweet group constipation efficiency was 95.24%,non-sweet group was 82.35%,and there was no significant difference between the two groups(P>0.05).5.Analysis on the correlation between the Momordica Grosvenor origin,fruit growth period,medicinal product specifications,and the relieving-cough expectorant and laxative efficacy.(1)The correlation between the origin of Siraitia grosvenorii and the relieving-cough,expectorant and laxative efficacy :From 37 batches of Siraitia grosvenorii of medium fruit,the fruit growth period was selected to be more than 85 days,and 19 samples of Siraitia grosvenorii from different origins were analyzed.The effective rate of relieving-cough of 19 batches of Siraitia grosvenorii was 84.21%.The coughing effect of Siraitia grosvenorii from different origins: The coughing effect of 2 batches Siraitia grosvenorii,which was produced in Lingui County,is 50%,which was somewhat lower.The difference efficacy of coughing of Siraitia grosvenorii in Lingui County between other origins was not significant.The effective rate of expectorant of 19 batches of Siraitia grosvenorii was 68.21%.The expectorant effect of Siraitia grosvenorii from different origins: The expectorant effect of 2 batches of Siraitia grosvenorii,which was produced in Lingui County,is 0%,the efficacy of expectorant of Siraitia grosvenorii from other origins is not much different.The effective rate of constipation of 19 batches of Siraitia grosvenorii was 84.21%.The laxative effect of Siraitia grosvenorii from different origins: The laxative effect of 2 batches of Siraitia grosvenorii,which was produced in Xingan County,is 0%,the efficacy of laxative of Siraitia grosvenorii from other origins is not much different.(2)The correlation between the growth period of Siraitia grosvenorii and the efficacy of relieving-cough,expectorant and laxative:From the analysis of relieving-cough test results,with immature fruit growth period of 40-45 days,there is no cough efficiency(R < 130%);with basically mature fruit growth period of 60-70 days,the coughing efficiency was 50%;with mature fruit growth period of 85-90 days,the coughing efficiency was 86.45%.It was suggested that the relieving-cough effect will also increase with the extension of the growth period of Siraitia grosvenorii.From the analysis of expectorant test results,with immature fruit growth period of 40-45 days and basically mature fruit growth period of 60-70 days,the expectorant efficiency was 100%(P < 0.05);with mature fruit growth period of 85-90 days,the expectorant efficiency was 61.29%.Tip With the extension of the growing period of Siraitia grosvenorii fruit,the expectorant effect is also weakened.From the analysis of laxative test results,with immature fruit growth period of 40-45 days and basically mature fruit growth period of 60-70 days,the laxative efficiency was 100%(P < 0.05);with mature fruit growth period of 85-90 days,the laxative efficiency was 87.09%.Tip With the extension of the growth period of Siraitia grosvenorii,the laxative effect is also weakened.(3)The correlation between the specifications of Siraitia grosvenorii and the efficacy of relieving-cough,expectorant and laxative:From 37 batches of Siraitia grosvenorii,those growth period with more than 85 days were selected,of which,9 batches from Longjiang Country,Yongfu County(1 large,1 small,7 medium);5 batches from Rongjiang Country,Xingan County(2 large,1 small,2 medium).The effective rate of relieving-cough for 14 batches of Siraitia grosvenorii was 92.85%.For 3 batches of big fruit,the relieving-cough rate was 66.67%,which is a little bit lower;for 9 batches of medium fruit,the relieving-cough rate was 77.78%;for 2 batches of small fruit,the relieving-cough rate was 100%.It is suggested that the large,medium and small Siraitia grosvenorii all have the efficacy of relieving-cough.The effective rate of expectorant for 14 batches of Siraitia grosvenorii was 57.14%.For 3 batches of big fruit,the expectorant rate was 66.67%;for 9 batches of medium fruit,the expectorant rate was 66.67%;for 2 batches of small fruit,the expectorant rate was 0%.It is suggested that there is a significant difference in the expectorant efficacy of different medicinal product specifications of Siraitia grosvenorii.The effective rate of laxative for 14 batches of Siraitia grosvenorii was 85.71%.For 3 batches of big fruit,the laxative rate was 100%;for 9 batches of medium fruit,the laxative rate was 77.78%;for 2 batches of small fruit,the laxative rate was 100%.It is suggested that there is no significant difference in the laxative efficacy of different medicinal product specifications of Siraitia grosvenorii.Conclusion:1.Siraitia grosvenorii does have significant relieving-cough,expectorant and laxative effects,which is consistent with the efficacy and indications of Siraitia grosvenorii regulated by the Chinese Pharmacopoeia(2015 version).And the experimental results show that Momordica Grosvenor laxative effect is stronger than cough and expectorant effect.2.There was a significant correlation between the content of Mogroside V and its relieving-cough efficacy,indicating that the content of Mogroside V described in Chinese Pharmacopoeia(2015 edition)under the Siraitia grosvenori quality was not less than 0.50%.The content of Mogroside V had no significant correlation with the efficacy of expectorant and laxatives,suggesting that Siraitia grosvenori may have other ingredients in sputum and laxative.3.Momordica Grosvenor fruit color dark or light,sweet or non-sweet taste,and its relieving-cough,expectorant and laxative efficacy had no significant correlation.It shows that the color and taste of Siraitia grosvenori are not the main factors affecting its efficacy.4.Siraitia grosvenorii from different place of origins may have little effect on cough,sputum and constipation.5.Siraitia grosvenorii with different fruit growth period may have little effect on its expectorant and laxatives,but it may affect the relieving-cough effect.With the extension of the growth period of Siraitia grosvenorii fruit,the relieving-cough effect was enhanced,but the expectorant effect was weakened and the laxatives effect was also weakened.It is suggested that the mothball harvest period should be 75-80 days for fruit growth period to ensure better relieving-cough,expectorant and laxatives efficacy.6.Siraitia grosvenorii,large,medium and small in different medicinal product specifications,may have little effect on relieving-cough and laxative,but it perhaps affect the expectorant effect.
