| ObjectiveTo explore the mechanism of Gua Sha on the rat model of lumbar disc herniation via two-dimensional electrophoresis.MethodsA total of 36 SD rats wew randomly divided into 6 groups,including Model Group(MO1 MO2 M03)and Gua Sha Group(GS1 GS2 GS3).Each group had 6 rats.A non-compression lumbar disc herniation model induced by autologous nucleus pulposus transplantation.Each rat underwnt an operation.Gua Sha Group was performed on the 5th day after the surgery,once every other day.3times is one period,and 1-3 courses of treatment according to groups.Model group only grabbed but did not scrapped.At the end of each period,the peripheral blood and the dorsal root ganglion in the corresponding group of rats were collected.Two-dimensional electrophoresis combined with mass spectrometry was used to explore the differentially expressed proteins related to the Gua Sha intervention and to analyze it by bioinformatics.Results(1)The expression of differential protein in serum①A total of 214 different proteins were identified between group GS1 and group MO1 A total of 166 different proteins were identified between group GS2 and group M02.A total of 225 different proteins were identified between group GS3 and group M03.There are 14 differential proteins both in the three periods,1 expression up-regulated and 13 down-regulation.②A total of 14 Gua Sha related proteins using GO analysis:7 molecular function,12 cellular component and 12 biological process were identified.In molecular function,ranked the top three,respectively were Binding(44%),Molecular function regulator(17%),Signal transducer activity(11%);In cellular component,ranked the top three,respectively were Organelle(20%),Cell part(20%),Extracellular region part(11%);In biological process,ranked the top three,Cellular process(20%),Biological regulation(20%),Response to stimulus(14%).③A total of 14 Gua Sha related proteins via IP A analysis.The top five canonical pathway were T Helper Cell Differentiation,LXR/RXR Activation,FXR/RXR Activation,Acute Phase Response Signaling,Role of IL-17A in Psoriasis.(2)The expression of differential protein in DRG①A total of 290 different proteins were identified between group GS1 and group MO1.A total of 350 different proteins were identified between group GS2 and group M02.A total of 323different proteins were identified between group GS3 and group M03.There are 27 differential proteins both in the three periods,13 expression up-regulated and 14 down-regulation.②A total of 27 Gua Sha related proteins using GO analysis:7 molecular function,10 cellular component and 18 biological process were identified.In molecular function,ranked the top three,respectively were Binding(41%),Catalytic activity(26%),Molecular function regulator(15%);In cellular component,ranked the top three,respectively were Organelle(29%),Cell part(29%),Membrane(13%);In biological process,ranked the top three,respectively were Cellular process(15%),Biological regulation(12%),Metabolic process(12%).③A total of 27 Gua Sha related proteins via IPA analysis.The top five canonical pathway were Chronic Myeloid Leukemia Signaling,Colorectal Cancer Metastasis Signaling,LXR/RXR Activation,HMGB1 Signaling,Aryl Hydrocarbon Receptor Signaling.Conclusions1.Gua Sha is involved in the treatment of LDH through multi-target,multi-channel and multi-channel.2.The mechanism of Gua Sha therapy for LDH includes not only the regulation of immune inflammation,but also the regulation of physiological metabolism.3.IL-17,TNF-a and NFκB1 may serve as potential targets,providing new ideas for explaining the mechanism of Gua Sha therapy for LDH. |
