Effect And Mechanism Of CRAE On Pulmonary Fibrosis Induced By Paraquat In Rats | | Posted on:2019-02-20 | Degree:Master | Type:Thesis | | Country:China | Candidate:Q F Guo | Full Text:PDF | | GTID:2394330545461433 | Subject:Emergency Medicine | | Abstract/Summary: | PDF Full Text Request | | Background : The theory of cytokine network imbalance plays an important role in the pathogenesis of acute paraquat poisoning.TGF-β1 is recognized as the initiating factor among many cytokines,Studies have found that epithelial and mesenchymal cells can be observed from morphological changes and TGF-β1 content increased obviously while stimulate A549 cells with paraquat.This results were confirmed that the TGF-β1 may be the key factor in the process of paraquat induced pulmonary fibrosis.In the development of pulmonary fibrosis,the expression of the signature protein of epithelial cells and the signature protein of mesenchymal cells can serve as an important basis for the identification of epithelial mesenchymal transformation(EMT).Traditional Chinese medicine(TCM)has been used in the treatment of lung diseases for thousands of years and it has rich experience and theoretical foundation.Astragaloside can adjust the metabolism of nitric oxide and strengthen cell anti-oxidation ability,so as to achieve the aim of protecting fibrosis of lung tissue.radix paeoniae rubra can reduce the generation and release of nitric oxide,thus alleviate peroxidation damage of lung tissue.So group according to the local "lung disease" folk prescription treatment combined with western medicine and traditional Chinese medicine theory and pulmonary fibrosis related basic research.Using waterextractmethod to preparation of Compound Radix Paeoniae Rubra Astragalus extract(CRAE).Giving different doses of CRAE to treat the poisoned rats after given paraquat solution to fill the stomach of rats to observe the change of various cytokines concentrations in the lung tissue and the change of the pulmonary fibrosis signature chyournel protein.From this way to understand CRAE its curative effect on pulmonary fibrosis caused by paraquat.Objective:To evaluate the effect and mechanism of the Compound of Radix Paeoniae Rubra and Astragalus extract on pulmonary fibrosis induced by paraquat in rats.Methods:The model of pulmonary fibrosis induced by paraquat was used to detect the effects of CRAE on lung weight、lung organ coefficient、HA、PC-Ⅲ、LN、MMP-9、TIMP-1、Hyp、MDA、SOD、GSH-Px、TNF-α、IL-17、IL-6、TGF-β1、PAI-1 in rats serum and tissues.The protein and m RNA expression of P-Smad2 、 Smad2 、PSmad3 、 Smad3 、 Smad7 and PAI-1 were detected by western blot and Q-PCR to evaluate the protective effect of CRAE.Results:CRAE(1.2 g/kg and 2.4 g/kg)could significant decrease lung weight、lung organ coefficient、HA、PC-Ⅲ、LN、TIMP-1、Hyp、MDA、TNF-α、IL-17、IL-6、TGF-β1、PAI-1 in rat rats serum and tissues,and increase the content of MMP-9、SOD and GSH-Px in rats tissues.In addition,CRAE(1.2 g/kg and 2.4 g/kg)could reduce the protein of P-Smad2 、 P-Smad3 、 PAI-1expression and increase the expression of Smad7.Conclusion:CRAE could improve the prognosis of pulmonary fibrosis induced by paraquat in rats and its mechanism might be related to TGF-β/Smad pathway. | | Keywords/Search Tags: | Compound of Radix Paeoniae Rubra and Astragalus extract, pulmonary fibrosis, paraquat, TGF-β/Smad pathway, PAI-1 | PDF Full Text Request | Related items |
