| Purpose We aim to investigate the effect of metformin butyrate(MFB)onhuman lung adenocarcinoma cell(A549)with different concentrations and different durations of intervention.We also explore the effect of MFB onthe expression of pigment epithelium-derived factor(PEDF).Methods After cell culture,according to different processing conditions,the cells were divided into control group(without drug intervention)and the experimental group(intervened by MFB concentrations with 0.5,1,2,4,8 mmol/L).For experimental group,MFB was intervened attwo durations(24 h,48 h)with different concentration.After intervention,the inhibition rate of MFB on A549 cell proliferation and the apoptosis rate were measured with MTT assay andflow cytometry,respectively.After intervention with MFB with different concentrations(0,2,4,and 8 mmol/L)for two durations(24 h 48 h),the migration of A549 cells was measured by scratch healing test.The level of pigment epithelium-derived factor(PEDF)in the culture supernatant of lung adenocarcinoma A549 cells were also measured by ELISA after MFB intervention with different concentrations(0,1,2,4 mmol/L)of MFB for 48 h.Finally,we compared the statistical differenceof groups on the levelof cell proliferation inhibition rate,apoptotic rate,migration ability,and protein expression of PEDF.Results The inhibitory rate of cell proliferation was significantly different between the control group and each experimental group(P<0.05).At the same duration of intervention,the inhibitory rate of proliferationwas greater in the high-concentrationintervention group thanthe low-concentration group.The experimental group of each concentration was significantly greater than the control group(P<0.05).At the same intervention concentration,the increase inhibition rate was significantly greater in groups with intervention duration of 48 h than 24h(P< 0.05).At the same duration of intervention,the apoptosis rate of the experimental group was significantly greater than that of the control group,and the apoptosis rate of the high concentration intervention group was significantly greater than that in the low concentration group(P< 0.05).At the same intervention concentration,the apoptosis rate was significantly greater in groups with intervention duration of 48 h than 24h(P< 0.05).The results of the scratch healing experiment indicated that under same duration of intervention,the migration ability was significantly smaller for the experimental group than the control group,and the high concentration group was significantly less than the low concentration group(P<0.05).At the same concentration,the migration ability at 48 hours was significantly less than 24 hours(P<0.05).The results of ELISA suggested that the level of PEDF protein was significantly higher in the experimental group than the control group and was significantly higher in the high concentration intervention group than the low concentration group(P<0.05).Conclusion MFB can inhibit the proliferation of human lung adenocarcinoma A549 cells in vitro with concentration-and time-dependentmanners.Meanwhile,MFB can induce apoptosis of A549 cells and inhibit the migration of A549 cells.Besides,the content of PEDF protein in the culture supernatant was positively associated with the concentration of MFB. |
