| Objective:To explore the effect of self-made Shuizhi Huoluo decoction on neurological recovery and brain protection in ischemic stroke mouse model.Methods:(1)production of ischemic stroke model:using photochemical method to make a mouse model of ischemic stroke.(2)Grouping and index measurement:In addition to the normal group,the remaining successful model mice were divided into model group,low dose(2g/kg)group,high dose(4g/kg)group,and aspirin(15mg/kg)group.Continuous administration for 28 days,in which the model group and the normal group were given distilled water(0.2ml/10g)for gavage.At the 1st,7th,14th,21st,and 28th days after modeling,each group was scored for behavior,rotarod fatigue test,and grip test.At last,blood samples were collected from eyeballs and the mice were sacrificed by cervical dislocation.The brains were made to a thickness of 2 cm.TTC dyeing,calculate the necrosis area in the brain,Blood samples were taken after centrifugation to measure the antioxidant index(T-SOD,MDA,NO).Result:(1)Behavioral score:at the 21st day after administration,,compared with the model group,the high-dose group and the aspirin group significantly increased(P<0.05);at the 28th day after administration,Compared with the model group,the high dose group and aspirin group increased significantly(P<0.05),compared with the low dose group,the aspirin group was significantly increased(P<0.05).(2)Measurement of rod fatigue tester:After 7 days of administration,the time of rotation of the aspirin group was significantly higher than that of the model group(P<0.05),compared with the low-dose group,the rotation time of the aspirin group was significantly increased(P<0.05);after 14 days,21 days,28 days of administration,the time of rotation were all prolonged(P<0.05,p<0.01).(3)Measurement of paw force:At 24 hours after operation,compared with the normal group’s grip test,the holding power of the model group,the low-dose group,the high-dose group,and the aspirin group was significantly reduced(p<0.01),indicating successful model establishment;at 7 days after dosing,the holding power of the model group,the low-dose group,the high-dose group,and the aspirin group was significantly lower than that of the normal group’s grasp test(p<0.01),compared with the grip test of the model group,the holding power of the low-dose group,the high-dose group,and the aspirin group was significantly increased(p<0.01),compared with the low-dose group’s grip test.Both the high-dose group and the aspirin group showed a significant increase in the holding power(p<0.01),after 14 days,21 days,28 days of administration,the grip test of each group of mice showed a tendency of increasing(p<0.01).(4)T-SOD:Test results showed that compared with the normal group,the test results of the model group were significantly reduced,the difference was statistically significant(p<0.01),compared with the model group,the results of the low-dose group,the high dose group and the aspirin group increased significantly(p<0.01),compared with the low-dose group,the results of the high-dose group and the aspirin group were significantly increased(p<0.01);MDA:The test results showed that compared with the normal group,the test results of the model group were significantly increased(p<0.01),compared with the model group,the test results of the high-dose group and the aspirin group were significantly reduced(p<0.01),and the low-dose compared with the group,the test results of the high-dose group and the aspirin group were significantly reduced(p<0.01);NO:The test results showed that compared with the normal group,the test results of the model group were significantly increased(p<0.01),compared with the model group,the test results of the low-dose group,high-dose group,and aspirin group were significantly reduced(p<0.01),compared with low-dose group,the test results in high-dose group and aspirin group were all reduced(p<0.01,p<0.05).(5)Cerebral infarct size:The area of cerebral infarction in the normal group was 0.Compared with the other groups and the model group,the results of low-dose group,high-dose group and aspirin group were significantly reduced,(p<0.01);compared with low-dose group,the results of high-dose group and aspirin group were significantly reduced(p<0.01).Conclusion:Self-made Shuizhi Huoluo decoction can significantly improve the degree of cerebral nerve damage in ischemic stroke mice,increase the activity of T-SOD in serum of mice,decrease the content of MDA and NO in serum to achieve brain protection,and can reduce the Rat brain ischemic area of death. |
