Study On OXA Gene And The Effect Of ISAba1 To Bla_oxa-23 In 90 Acinetobacter Baumanii

Objective:To study the the Yijishan Hospital of Wannan Medical College about OXA gene popular trend,as well as the effect of ISAba1 to bla_oxa-23xa-23 in Acinetobacter baumanii.Methods:Using VITEK 2 compact automatic microbe testing of 90 non-replicate Acinetobacter baumannii strains,multiple polymerase chain reaction（MPCR）to detect four OXA genes and inserted sequence1（ISAba1）in Acinetobacter baumanii.TA cloning resistant gene sequence blaoxa-23,insertion sequence ISAba1 and ISAba1-blaoxa-23,carring on the transformation with E.coli,The minimal inhibitory concentrations（MIC）of the positive strains and the products of the transformation to imipenem and meropenem were determined using broth dilution.Results:Multiple polymerase chain reaction detect of 90 non-replicate Acinetobacter baumannii strains,73 of blaoxa-23are positive strains,90 of blaoxa-51 are positive strains,86 of ISAba1 are positive strains,not detected of blaoxa-24 and blaoxa-58positive strains.17 of blaoxa-23 negative strain has 11strains are sensitive to imipenem,in which 11 strains of four strains are not detected ISAba1.The MIC of positive strains of pET28b（+）-blaoxa-23、pET28b（+）-ISAba1、 pET28b（+）-ISAba1-blaoxa-23 and transformant（pET28b（+））to imipenem are 4ug/ml,1ug/ml,64ug/ml and 0.12 ug/ml,the meropenem are 16ug/ml,4ug/ml,0.5ug/ml and 0.03ug/ml respectively.Results show that the ISAba1 affect blaoxa-23 gene expression.Conclusion:The south of Anhui province popular gene are blaoxa-23 and blaoxa-51.None of them are sensitive,Both of them are high drug resistance.This situation has been confirmed that the Acinetobacter baumanii drug resistant are high related of blaoxa-23 and ISAba1.ISAba1 can result high expression to blaoxa-23 may be carbapenems antibiotics highly resistant main reasons in Acinetobacter baumanii.
