In order to study the response of growth and physiological and biochemical characteristics of potato tissue culture seedlings to photoperiod treatment,and the photoperiod sensitivity of potato,three photoperiod treatments were designed.Three potato varieties(‘foreign 2',‘Minshu 1' and ‘Qingshu 9')were used as experimental materials.The phenotypic characteristics(plant height,stem diameter,stem node number,leaf number,root length,root number and fresh weight)of potato were analyzed,dry weight)and physiological and biochemical indexes(soluble protein,catalase,superoxide dismutase,proline,malondialdehyde),the effect of photoperiod treatment on Physiological Indexes of different varieties,the index characters that can reflect the photoperiod sensitivity of potato were analyzed and screened,and the relative photoperiod sensitivity of three potato varieties was analyzed.Stco gene was cloned from ‘Qingshu 9'by homologous cloning technology:1.Through the comprehensive analysis of eight growth indexes,such as leaf number,fresh weight,stem diameter,stem node number,root number,dry weight,root length and plant height,it is concluded that the comprehensive growth performance of ‘foreign 2' and ‘Minshu 1'.2.16h/d light treatment can significantly promote the synthesis of soluble protein content of ‘foreign 2' potato,and 8h/d light treatment can significantly promote the synthesis of soluble protein content of ‘Minshu 1' and ‘Qingshu 9'potato.The activities of SOD and cat did not show a certain regularity due to the fact that the two kinds of oxidase of ‘foreign 2' and ‘Qingshu 9'were not sensitive to photoperiod.The activity of SOD was the highest and the accumulation of MDA was the least under 12h/d light.3.The fresh weight,dry weight,stem diameter and root length of the three potato varieties increased with the increase of light hours.4.Variance analysis showed that photoperiod had significant effects on dry weight,number of roots,fresh weight and total root length of potato(P<0.05);varieties had significant differences on plant height and number of roots(P<0.01);interaction effects of photoperiod and varieties also had significant effects on stem diameter,solubleprotein,catalase(CAT)superoxide dismutase(SOD),malondialdehyde(MDA)and proline(pro)Significant effect(P<0.01).5.Four phenotypic traits(dry weight,fresh weight,number of roots,length of roots and so on)were selected as the relative sensitivity index of photoperiod.The results show that ‘Qingshu 9'is less sensitive to photoperiod than Minshu 1 and ‘foreign 2'.6.In this study,stco gene was cloned from ‘Qingshu 9'.It encodes 413 amino acids and has a total length of 1242 BP.The protein encoded by stco gene is composed of irregular curl(Cc),?-helix(Hh),elongating ligation(Ee)and ?-folding(Tt).The highest proportion of irregular curl is 57.87%,and the proportion of ?-folding(Tt)is 1.69%.7.The results of fluorescence quantitative PCR showed that stco in ‘Qingshu 9' In ‘foreign 2'and ‘Minshu 1' the expression of ‘Qingshu 9'was lower than that of ‘foreign 2'and ‘Minshu 1'under three simulated photoperiod conditions.
