"Sanghuang" is a well-known medicinal fungus in China."Sanghuang",which has many effects,has been recorded in many Chinese herbal books in ancient China.In this study,five strains of "sanghuang" were collected to study the solid and liquid culture characteristics of their mycelia,the bottle cultivation method was used to cultivate "sanghuang" with different grain culture media,the changes regulation of flavonoids in different growth stages of "sanghuang" was discussed,and the diversity of flavonoids of 5 strains cultures was analyzed.First at all,the author collected and identified five different strains through multiple channels in this study: Sanghuangporus quercicola,Phellinus nigricans,Tropicoporus linteus,Sanghuangporus bamuii,Inonotus hispidus.In this study,the solid culture characteristics of five strains were analyzed from three aspects: basic medium,carbon source and nitrogen source.It shows that the most suitable basal medium for Sanghuangporus quercicola,Phellinus nigricans,Tropicoporus linteus,Inonotus hispidus is PDA medium,and the most suitable basal medium for Sanghuangporus bamuii is GPY medium.The carbon sources suitable for the growth of five strains include mannose,at the same time,fructose and glucose are also suitable for the growth of multiple strains.The nitrogen sources,which all include peptone,are suitable for the five strains to grow.In addition,none of the five strains could grow normally on the medium with urea.In this study,the liquid culture characteristics of five strains were analyzed from three aspects: basic medium,carbon source and nitrogen source.It shows that the most suitable basal medium for Sanghuangporus quercicola,Sanghuangporus bamuii and Inonotus hispidus is GPY liquid medium,the most suitable basal medium for Phellinus nigricans is PDA liquid medium,the most suitable basal medium for Tropicoporus linteus is wheat bran liquid medium.In liquid culture,the suitable carbon source of Sanghuangporus quercicola,Inonotus hispidus is fructose and the suitable nitrogen source is peptone;the suitable carbon sources of Phellinus nigricans are xylitol and sorbitol and the suitable nitrogen source is beef paste;Mannose and peptone,which are carbon and nitrogen sources respectively,are the most suitable ones for Tropicoporus Iinteus;at the same time,for Sanghuangporus bamuii,glucose and beef paste are the best.The study explored the bottle culture technology of "sanghuang" fruiting body.During the cultivation process,multiple strains produced black droplets on the surface of oat and semen coicis medium,with an irritating odor and the mediums were soft and rotten.About Tropicoporus linteus on millet medium,and Sanghuangporus bamuii on rice and millet medium,soft and rotten mediums also appeared.Throughout the cultivation process,Sanghuangporus quercicola,Phellinus nigricans,Tropicoporus linteus,Sanghuangporus bamuii grew well on corn medium.Inonotus hispidus did not form fruiting bodies on the rice and corn medium,and grew well on the remaining three medium.This study analyzed the changes regulation of the flavonoids in different growth stages of five strains.In the later stages of cultivation,the total flavonoid yield of Sanghuangporus quercicola,Phellinus nigricans,Tropicoporus linteus,Sanghuangporus bamuii on corn medium was higher than that of other experimental medium.Inonotus hispidus had lower total flavonoid production on millet,oat,and coix seed medium,all of which did not exceed 1 mg/g.Among the five strains,the total flavonoid yield of Phellinus nigricans on the corn medium was the highest,with a maximum yield of 6.41 mg/g.In this study,in order to explore the diversity of flavonoids in five kinds of strains,a total of 10 flavonoids metabolites were detected,six kinds of flavonoids were detected in Sanghuangporus quercicola,Phellinus nigricans,Tropicoporus linteus,Inonotus hispidus,eight kinds of flavonoids were detected in Sanghuangporus bamuii.The five strain cultures all contained the five compounds of Catechin,Di-O-methylquercetin,Tangeretin,Nobiletin,3,5,6,7,8,3’,4’-Heptamethoxyflavone,among them,the relative content of 3,7-dioxo-methylquercetin is generally higher.
