The Establishment And Evaluation Of Liver Injury Model Of Procambarus Clarkii | | Posted on:2021-05-16 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y N Wu | Full Text:PDF | | GTID:2393330611461471 | Subject:Aquaculture | | Abstract/Summary: | PDF Full Text Request | | With the booming market demand for Procambarus clarkii in China in recent years,many areas have expanded the scale of the cultivation of Procambarus clarkii.With the paddy fields and other waters,pesticides usually enter the water environment through the soil.Hepatopanax,as an important immune and detoxifying organ of crustaceans,is often damaged by the toxic effects of pesticides,thus affecting the growth of procambarus crustaceans.In this study,the hepatopancreas injury of Procambarus clarkii was induced by high efficiency cypermethrin,and the appropriate time and drug concentration for the establishment of the hepatopancreas injury model of Procambarus clarkii were explored.To study the safety and effective dose of turmeric powder on hepatopancreas of Procambarus clarkii.This study laid a foundation for the future study of the occurrence,development and pathological characteristics of hepatopancreatic injury,and provided theoretical basis for the evaluation of the efficacy of prevention.Specific studies are as follows:1.The method of inducing hepatopancreatic injury model of Procambarus clarkii with high efficiency cypermethrinProcambarus crustaceans were infected with highly effective cypermethrin solution with gradient concentration,and samples were collected at each time point for detection and observation.The results showed that compared with the control group,the serum transaminase activity of the 0.02 regimen group was significantly increased(P<0.05);TP(total serum protein)was significantly different from SOD(superoxide dismutase),CAT(catalase)and MDA(malondialdehyde);The relative expression of Pc Toll-receptor m RNA was down-regulated in hepatopancreatine injury model.The results of pathological sections showed that cypermethrin was infected for three days,and the liver and pancreas showed obvious inflammatory reaction.The results showed that the hepatopancreas injury model could be established by soaking with 0.02 g/L cypermethrin in 3d.Serum biochemical indexes ALT,AST,TP andhepatopancreatinase SOD,CAT and oxidation product MDA can be used as indexes to judge the degree of hepatopancreatinosis injury,but ALP(alkaline phosphatase)and GLU(blood glucose)are not suitable as indexes.2.To study the safety and dose of turmeric powder on procambarus crustaceansProcambarus clarkii were fed with different doses of turmeric powder for 30 days,and then the samples were collected for testing.The results showed that compared with the blank control group,the 0.2g/kg dose group and the 0.6g/kg dose group,there was no significant difference in the number of survival of Procambarus clarkii in blank control group(P>0.05),survival was significantly reduced in the1g/kg group(P<0.05);Biochemical indicators in the 0.2g/kg and 0.6g/kg groups showed no significant changes compared with the blank control group(P>0.05),TP was significantly reduced in the 1g/kg dose group(P<0.05).THC(total blood cell count)and DHC(different types of blood cell count)were significantly reduced in.1g/kg hepatopancreatine sections showed mild inflammatory response.Conclusion:0.2g/kg dose baits and 0.6g/kg dose baits can be used for liver protection test.3.To study the effective dose of turmeric powder for hepatopancreatic injury repairThe experimental shrimp with induced liver injury were randomly divided into 5groups.The samples were analyzed after feeding the bait continuously for 10 d.The results showed that the activity of CAT and SOD increased with the increase of the dose.CAT activity and SOD activity in the 0.4g/kg dose group were significantly increased compared with the negative control group(P<0.05),the content of MDA was the least in the 0.4g/kg dose group.The relative expression of Pc Toll-receptor gene in hepatopancreatine was significantly increased after 0.4g/kg dose of Procambarus clarkii was fed for 10d(P<0.05).The pathological results showed that the cells in the 0.4g/kg group were closely arranged,with normal cell size,fewer fat vesicles in the cells,and a good recovery of hepatopancreatine.The above results showed that: 0.1g/kg~0.4g/kg of turmeric powder had a certain protective effect on hepatopancreas of Procambarus clarkii,and the protective effect was in a dose-effect relationship with the dose.The dose of 0.4g/kg of turmeric powder could repair the hepatopancreas and improve the immune function of Procambarus clarkii. | | Keywords/Search Tags: | beta-cypermethrin, Procambarus clarkii, liver injury, turmeric powder, PcToll receptor | PDF Full Text Request | Related items |
