Mannose oligosaccharide(MOS)is supplied in feeds as additive to promote the growth performance of animals,improve the intestinal digestion environment,and enhance the intestinal mucosal immune function.This research was focused on establishing the intestinal injury model of rats which induced by aspirin(ASA),and investigated the effect of inhibition on intestinal injury in rats by MOS,then discussed MOS repair mechanism to damage intestinal in rats.(1)Establishment of the model of ASA-induced intestinal injury in rats.The single-factor experiment design was adopted in the experiment.Different doses of ASA were administered to 6-week-old SD rats and intestinal injury was observed.The results show that the rat body weight,serum interleukin 2(IL-2),the intestinal mucosa secretory immunoglobulin A(sIgA),the intestinal villus height(VH)and the ratio of villus height to crypt depth(CD)were reduced significantly by 50 mg/kg aspirin via oral administration compared with the control group(P < 0.05),but there were no effects on the liver index,spleen index and the activity of serum lysozyme(LZM)(P > 0.05).200 mg/kg aspirin was administrated to rats can significantly reduce the body weight gain,liver index,serum IL-2 content and LZM activity,intestinal mucosal sIgA content,intestinal villi height and crypt depth,and the ratio between them(P < 0.05).In this study,the rat model of intestinal injury was successfully established by oral administration of 200 mg/kg aspirin for 2 weeks.(2)Study on the mechanism of MOS inhibiting intestinal injury in rats: A single factor design,36 6-week-old SD rats were given to 6 treatments(blank group,ASA group,100,300,600 and 800 mg/kg MOS groups)with 6 replicates,1 rat in each replicate(single cage feeding).Compared with the ASA group,600 and 800 mg/kg MOS improved the liver index,serum IL-2 and LZM content,sIgA and mucus volume,VH,CD and VH/CD of the immune injured rats(P < 0.05).An appropriate dose of MOS can improve the immune ability of rats and inhibit the intestinal injured.(3)Study on the repair mechanism of MOS to ASA-induced damage intestinal in rats: A single factor experiment design,36 6-week-old SD rats were allotted to 6 treatments(blank group,ASA group,MOS group,and 50,100 and 200 mg/kg ASA with 600 mg/kg MOS groups)with 6 replicates,1 rat in each replicate(single cage feeding).Results show that compared with ASA group,50 mg/kg ASA group in the serum IL-2,LZM,total superoxide dismutase(T-SOD)CD and VH/CD did not exist significant difference(P > 0.05),for 100 mg/kg ASA group,there is no significant difference in the serum IL-2,CD and VH/CD with the blank group(P > 0.05),and 200 mg/kg ASA group T-SOD increased significantly in the model group(P < 0.05),but with the blank group(P > 0.05),there are still differences However,VH/CD was significantly higher than that of the model group(P < 0.05),and was not different from that of the blank group(P > 0.05).Under the present experimental conditions,600 mg/kg of mannooligosaccharide had a good repair effect on low-concentration aspirin-induced intestinal injury in rats.
