ATG26 has been identified as a factor that activates peroxisome autophagy on Pichia pastoris.It was found in anthracnose that the absence of ATG26 results in a slower growth rate of ΔCo ATG26 on the PDA medium and reduced pathogenicity.However,the function of ATG26 in Fusarium graminearum is unclear.In this study,we first knocked out the FgATG26 gene by homologous recombination to obtaint ΔFgATG26,and backfilled to obtain the complementΔFgATG26-C.ΔFgATG26 showed reduced mycelial growth rate on the culture medium.ΔFgATG26 reduced conidia production.The sexual reproduction ofΔFgATG26 is affected,and the production of ascomycetes is reduced.ΔFgATG26 increased sensitivity to tebuconazole.Experiments were carried out by inoculating wild-type PH-1 and ΔFgATG26 in corn whiskers and field wheat ears.It was found that the ΔFgATG26 had significantly lower infectivity than the wild-type PH-1 in both the indoor test corn and field test wheat ears.The measurement of toxin content in wheat ears inoculated in the field showed that the DON toxin content in theΔFgATG26 was also significantly reduced compared to wild-type PH-1,indicating that the deletion of FgATG26 would seriously cause pathogenicity of Fusarium graminearum.By detecting the transcription level of the TRI gene cluster that regulates the key genes for Fusarium graminearum toxin production,it was found that in ΔFgATG26,the expression of TRI genes related to toxin production was significantly reduced.The above experimental results indicate that FgATG26 plays an important role in the growth and development,asexual reproduction,sexual reproduction and pathogenicity of Fusarium graminearum.Studying the function of FgATG26 can provide a theoretical basis for controlling The above experimental results indicate that FgATG26 plays an important role in the growth and development,asexual reproduction,sexual reproduction and pathogenicity of Fusarium graminearum.Studying the function of FgATG26 can provide a theoretical basis for controlling the wheat head scab.
