| Cold stress refers to the corresponding non-specific immune response of animals when they are stimulated by cold.The ability of an animal body to resist cold stress has been a focus for most researchers lately and the main focus has been on regulation of nutrition by addition feed additives,use of medicines as well as use of natural coldstress resistant-enhancing substance.Therefore,it is of great practical significance for the healthy growth of animal and animal husbandry to seek the substances with anticold stress performance from natural plants and to study the mechanism of their effects on the healthy growth of animals.The rhizomes of Acanthopanax senticosus are a potential plant source of such with Acanthopanax senticosus saponin B as a main chemical component.The hypothalamus controls thermoregulation and the main mechanism of cold stress is the co-regulation of SAM,HPA and HPT.In this study,we investigated the effects of Acanthopanax senticosus saponin B on cold stress on the hypothalamus of mice.For this study,mice were taken as the experimental animals.The effects of Acanthopanax senticosus saponin B on hypothalamus anti-cold stress were studied by establishing cold stress model,feeding experiment,slaughter test and molecular test.This provided theoretical basis for revealing the mechanism of cold stress on animal body and maintaining its normal physiological function.The protein expression of Hsp70 in hypothalamus of cold-stressed mice was detected by ELISA method.The optimum concentration and action time of Acanthopanax senticosus saponin B were also determined.Roche biochemical instrument was used to determine the changes of AST,ALT,LDL-CH,HDL-CH,TG,LDH,T3 in Serum;Detection of expression of Hsp70,ACTH,CRH,TRH protein and relative expression of mRNA in hypothalamus was done by Western-blot and fluorescence quantitative PCR to explore the effect of Acanthopanax senticosus saponin B on cold stress in hypothalamus of mice.The results are presented:(1)Under the conditions of this experiment,the injection concentration and action time of Acanthopanax senticosus saponin B were determined by Hsp70.4mg/kg Acanthopanax senticosus saponin B increased the protein expression of Hsp70.The successful establishment of cold stress model can be further used to study the mechanism of Acanthopanax senticosus saponin B on cold stress in mice.(2)The research on the effects of Acanthopanax senticosus saponin B on related blood indexes in cold-stressed mice showed that Acanthopanax senticosus saponin B could promote the ability of resisting cold stress condition.Acanthopanax senticosus saponin B significantly reduced the content of LDL-CH,the activity of ALT,AST,CK and LDH,but increased the contents of HDL-CH and TG.This enhanced the ability to resist cold stress by the hypothalamus of mice.(3)It was observed Acanthopanax senticosus saponin B can up-regulate the hypothalamic anti-cold stress related factors and hormones in mice,and can significantly increase the relative expression of Hsp70,ACTH,TRH,CRH(p< 0.05).In summary,this study found that 4 mg/kg Acanthopanax senticosus saponin B has protective effect on hypothalamus of cold stressed mice.This protective effect is to increase the content of ACTH,TRH,CRH and other stress hormones in the hypothalamus.This is achieved by inducing activation of SAM system,HPA axis and HPT axis,and to increase the expression of stress protein Hsp70 in hypothalamus of cold stressed mice,so as to enhance the anti-cold stress ability of hypothalamus. |
