| The tobacco aphid Myzus persicae(Sulzer)is a common pest on tobacco.It can cause great impact on the growth and development of tobacco,and also spreads a variety of plant viruses,has caused great economic losses to agriculture in China.The parasitoid Aphidius gifuensis Ashmaed is the endoparasite natural enemy of the tobacco aphid.The parasitoid is used to control the tobacco aphid,reduced the population of the tobacco aphid and the economic losses.The diversities of symbiont in the tobacco aphid and parasitoid in Fujian province were analyzed by the high-throughput sequencing of 16 Sr RNA and ITS region.The differences of species and diversities of symbiont were compared between male and female adult parasitoids,indoor and outdoor populations,different area populations.The results are as follows:(1)Based on the results of species classification,fungal communities of indoor population from aphids belonged to 7 phyla,19 classes,48 orders,87 families,125 genera and 170 species.Among them,the dominant population was Wallemia mellicola.Bacterial species of the tobacco aphid(indoor population)belonged to 29 phyla,59 classes,124 orders,242 families,517 genera and 804 species.The fungal communities of the tobacco aphid(outdoor population)belonged to 4 phyla,14 classes,31 orders,45 families,61 genera and 87 species.Among them,the dominant population was in Saccharomycetales.Bacterial species of the tobacco aphid(outdoor population)belonged to 26 phyla,54 classes,113 orders,187 families,393 genera and 585 species.The dominant species was in Bacillus.(2)Based on the results of species classification,fungal communities of indoor population from the female parasitoids belonged to 5 phyla,14 classes,24 orders,31 families,40 genera and 52 species.Among them,the dominant population was W.mellicola.Bacterial species of the female parasitoids(indoor population)belonged to 28 phyla,53 classes,106 orders,177 families,365 genera and 536 species.The dominant species was in Bacillus;The fungal communities of the male parasitoids(indoor population)belonged to 5 phyla,15 classes,27 orders,37 families,45 genera and 59 species.Among them,the dominant population was W.mellicola.Bacterial species of the male parasitoids(indoor population)belonged to 31 phyla,63 classes,136 orders,277 families,475 genera and 767 species.The dominant species were Ruminococcaceae_UCG-005.(3)Based on the results of species classification,fungal communities of outdoor population from the female parasitoids belonged to 6 phyla,17 classes,29 orders,38 families,49 genera and 68 species.Among them,the dominant population was in Saccharomycetales.Bacterial species of the female parasitoids(outdoor population)belonged to 19 phyla,34 classes,65 orders,108 families,253 genera and 397 species.The dominant species was in Bacillus;The fungal communities of the male parasitoids(outdoor population)belonged to 6 phyla,19 classes,35 orders,50 families,69 genera and 98 species.Among them,the dominant population was W.mellicola.Bacterial species of the male parasitoids(outdoor population)belonged to 34 phyla,64 classes,142 orders,248 families,496 genera and 742 species.The dominant species was in Bacillus.(4)Based on the analysis of the endosymbiotic diversity of adult parasitoids from different tobacco planting areas in Fujian,the bacterial communities of parasitoids belonged to 4 phyla,6 classes,11 orders,15 families,22 genera and 48 species.Wolbachia was detected in most of parasitoids from tobacco planting area in Fujian,and Rickettsia was found in almost all of parasitoids in Fujian.(5)The composition of symbiotic fungi in the outdoor aphid population was more abundant than that in the indoor population.The species of fungi in the indoor female parasitoids was less than that in the outdoor population,while the bacterial species in the indoor parasitoid population was more than those in the outdoor population.And the species of fungi in the indoor female population was obviously less than those in the outdoor population.The species of bacteria in the outdoor parasitoid population was not different from those of the outdoor population.The composition of symbiotic fungi and bacteria between the female and male parasitoids was the same as those in the indoor population.The species of symbiotic fungi in the female parasitoids was slightly more than those in the male parasitoids,and the species of symbiotic bacteria was obviously less than those in the male parasitoids.(6)The dominant symbiotic fungi in the indoor populations of M.perisicae and A.gifuensis was W.mellicola,and the dominant symbiotic fungus of the male species in the outdoor parasitoid population was also W.mellicola.The dominant symbiotic bacteria were Bacillus spp.not only in the outdoor aphid population but also in the female and male parasitoids from the outdoor population,and in the female parasitoids from the indoor population. |
