"New Flower Of Mid-Autumn Festival" Hemerocallis In Vitro Rapid Propagation Research

Hemerocallis is considered a good landscape and garden ornamentals for its strong plants,large flowers with bright color,elegant leaf shape and upright leaf posture.Hemerocallis of wild germplasm resources in China is extremely diverse,but the long-term natural and artificial hybridization has resulted in the similar origin of day lily,to some extent,complex genetic background bring difficulties to breeding work.In recent years,Chinese researchers have actively collected excellent germplasm resources and introduced many kinds of daylily varieties from abroad,to develop new varieties breeding research work.This research takes the lilies of the late flowering variety"new flower of Mid-Autumn festival"cultivated by Qidong Food co.,LTD as the research object,according to different time,different methods of sterilization were used for explants consisting of the parts,such as the flower bud,the upper part of the flower stalk,the leaf and the stems.This study used different concentrations of plant growth regulators for callus induction and adventitious bud to start training,at the end of the experiment,the adventitious buds were induced to proliferate,rooting and transplanting.The aim of the experiment is to find out the right way to establish the rapid propagation technique system of hemerocallis"new flower of Mid-Autumn festival",is aim to provide seedling basis for factory production,and provide technical support for hemerocallis new varieties deep breeding.The test results show that:（1）Considering pollution rate of explants,the rate of death and the degree of disinfection,this study consider the nodules of the young pedicle tiller as the best explant of"new flower of Mid-Autumn festival"hemerocallis tissue culture,the period of sampling is in the middle of August,and the sterile seedlings can be effectively obtained.The explants can sprout in about two weeks and form callus in a month,which shorten the time of obtaining sterile seedlings to a large extent.Taking young peduncle,the leaf and ovary as experimental material,the best sterilization solution is C2 treatment:70%Ethanol（C₂H₅OH）30 s+0.1%Mercury dichloride（HgCl₂）for 10 min（add right amount of Polysorbate）.Taking rhizomes as experimental material,the best sterilization solution is C9 treatment:70%Ethanol（C₂H₅OH）30 s+0.1%Mercury dichloride（add the right amount of Polysorbate）12 min+2%Sodium Hypochlorite（NaClO）4 min.（2）Through the influence result of the plant growth regulator on"new flower of Mid-Autumn festival"hemerocallis varieties germinating culture initiation rate of healing rate analyses that P7 treatment:MS+6–BA 1mg/L+NAA 0.1 mg/L,have better effect for the"New flower of Mid-Autumn festival"varieties of explant callus induction rate,the germination rate and promoting the explant morphogenesis.Therefore,this treatment is the best starter culture medium for the tissue culture of the pedicel,ovary and rhizomes of the hemerocallis.For the explants of the leaves,the best starter culture medium was EP:SH+Proline（C₅H₉NO₂）0.3g/L+Agarose（AG）4g/L+2,4-d 3.0mg/L+TDZ 0.5mg/L+Inositol100mg/L+Sucrose 30g/L.Though observe the influence result of ten treatments of the proliferative rate and proliferative coefficient,shows that suitable plant growth regulator concentration and type collocation can effectively promote the formation of plants,and improve the proliferation coefficient of tissue culture seedlings,and obtain healthy and excellent tissue culture seedlings.According to the experimental demonstration,the best growth medium for"new flower of Mid-Autumn festival"hemerocallis is Q7 treatment:MS+6-BA 2.0mg/L+NAA0.2mg/L.（3）The most suitable rooting medium for"New flower of Mid-Autumn festival"is Z6treatment:1/2MS+NAA 0.1mg/L+IBA 0.1mg/L.If you want to take root quickly,you can use 7 days rooting culture regardless of production cost:1/2MS+NAA 0.2mg/L,and then access to 1/2MS+NAA 0.1mg/L+IBA 0.1mg/L medium.There were differences in the growth and growth rate of the two treatments and no significant differences between the rooting rate and survival rate of transplanting.The number of seedlings with normal growth was more than 3,and the root length was more than 2cm.No matter which matrix is transplanted,the tissue culture can be viable under normal management.（4）It shouldn’t be more than 2 days before the hardening-seedling time of hemerocallis,and the suitable hardening-seedling time of"New flower of Mid-Autumn festival"hemerocallis is 1 day.According to the analysis results,the most suitable culture matrix of"New flower of Mid-Autumn festival"hemerocallis is V（Garden soil）:V（Vermiculite）=1:1 mixed matrix.In order to reduce the bacteria and germs in soil,the cultivation matrix should be treated with high temperature and high pressure sterilization before transplanting.In this way,the survival rate of the"new flower of Mid-Autumn festival"hemerocallis seed seedlings after transplanting could reach 100%,and all kinds of substrates have good growth potential.
