| In order to investigate the role and mechanism of iNOS in the lung tissue of yak at high altitude during hypoxia adaptation.The Yaks and cattle from different altitudes were selected as the experimental objects.Detecting the content of NO in arterial blood,comparing and analysing the relative content of iNOS mRNA and protein in cattle and yaks’ lung tissues.The relative abundances were detected by using quantitative real-time PCR and western bloting.The results are as follows:1、The content of NO in the blood of cattle showed an increasing trend at altitude,the content of NO in blood of Haiyan cattle was the highest,which was significantly higher than the Haiyan yak at the same altitude;the content of NO in blood of Datong yak was higher than the Haiyan yak at the same altitude,but the difference was not significant.2、The relative abundances of iNOS mRNA in the lung tissue of both cattle and yak showed an increasing trend of elevation,and the highest was the cattle in Haiyan;Qinchuan cattle were significantly higher than Qinchuan cattle in the plateau;Datong yak was higher than Haiyan yak at the same altitude,but the difference was not significant.3、The relative content of iNOS protein in lung tissue of cattle and yak showed an increasing trend of elevation,and Haiyan cattle was significantly higher than Haiyan yak;Qinchuan cattle were significantly higher than Qinchuan cattle in the plateau;Datong yak was significantly higher than Haiyan yak.The above results suggest that the expression of iNOS mRNA,protein and the content of NO are consistent,showed that the expression and regulation of iNOS mainly occurred at the transcription level;The expression of iNOS mRNA,protein in lung tissue,the content of NO in blood were influenced by altitude and variety;the expression of iNOS in lung tissue plays an important role in animal adaptation to hypoxic environment. |
