This experiment by means of radix aconiti(monkshood)two cultivated a growth and development characteristics in the whole stages,transcriptome sequencing analysis and metabolomics were performed on the harvest time subroots,and detection four kinds of alkaloid content in root of aconite monkshood processed products(such as black pills,white pills)and Ten kinds of proprietary Chinese medicine,Discuss differences metabolites in the important position in the application value of lateral root.Specific results are as follows:1.Liangshan cultivated species have bigger or higher leaves,plant height,stem weight,mother roots weight than Qingchuan cultivated species,Liangshan cultivated species the appearance of higher yield.2.According to the gene expression difference in 275 grown in two kinds of differentially expressed genes,including 58 genes rise significantly in Liangshan cultivated species,217 genes significantly lower,the function of these differentially expressed genes function mainly relate to the synthesis of cell wall and cell wall,cell wall structure,and the enrichment to KEGG pathways including Tryptophan metabolism degradation,Lysine degradation,Photosynthesis-capture optical antenna and Isoquinoline alkaloid synthesis pathway etc.3.Liangshan cultivated species and Qingchuan cultivated species cultivated species can clearly distinguish,prove that look from the chemical composition of two cultivated species does exist obvious differences;Screening get 40 different metabolites,some of these metabolites such as betaine,l-carnitine also has definite pharmacological activities,so they have a certain quality evaluation significance,they enrichment to KEGG pathways including Isoquinoline alkaloid synthesis pathway etc.4.Liangshan cultivated species three poisonous alkaloid is significantly higher than Qingchuan cultivated species;the content of DMC in Qingchuan cultivated species were significantly higher than Liangshan cultivated species;the content of four alkaloids in black pleces were higher than that in white pleces;Ten kinds of commercial proprietary Chinese medicine,the double ester content of total alkaloidsof 0-0.0644μg/g,content of DMC were 0.0044-0.5461μg/g,to prove to DMC is the key of monkshood,and DMC is the potential quality marker of monkshood.
