Comparison And Analysis Of Several Serological Detection Methods For Brucellosis

Brucellosis is a very harmful zoonotic disease.In recent years,the infection rate of brucellosis in China is on the rise.At present,there are many detection methods for Brucellosis in domestic and foreign markets,such as ELISA kit and colloidal gold test.Paper strips,etc.In this paper,10 kinds of reagents and kits produced by different manufacturers at home and abroad were used to test and compare the different among infect sheep serum,Brucellosis vaccine Rev.l,S2 immune sheep serum,unimmunized cattle and sheep positive serum,and negative.Serum was tested and compared to provide a basis for the selection of detection methods for Brucellosis in production.The results showed that,(1)For the virulent infection positive sheep serum,the sensitivity of G6,RBPT produced by G3,SAT reagents and G2,G5 colloidal gold is the best,and the sensitivity of each of the above is 100%.Besides in 4 ELISA methods,The sensitivity of G4c-ELIS A was 80%and the remain 3 are less sensitive.(2)For the known negative/positive bovine serum,the sensitivity of each detection method is:G3 RBPT is 97.83%,G5 i-ELISA is 95.65%,G4 b-ELISA is 93.48%,the sensitivity of G3 SAT,G2 i-ELISA,G2 c-ELISA was 75%,73.91%,and 76.09%respectively.The sensitivity of colloidal gold rapid detection card was 96.74%for G5,81.52%for G2,and 68.48%for G1.In addition to the specificity of G5 rapid test card is 93.33%,the specificity of the remain methods is 100%.(3)For the known negative/positive sheep serum,the sensitivity of G6,G3 RBPT is highest,and both are 98.33%;the sensitivity of G6,G3 SAT is 91.67%and 93.33%respectively;the sensitivity of G4b-ELISA,G5 c-ELISA is 85.83%and 81.67%.The sensitivity of three methods of G2,i-ELISA,c-ELISA and colloidal gold is 63.33%,43.33%and 67.5%,respectively.The specificity of G3RBPT and G4b-ELSIA is 99.17%,and the specificity of remaining method is 100%.(4)The sensitivity of G6 RBPT was the highest in the positive serum of Rev.1,which was 97.7%.The sensitivity of G4 b-ELISA was the highest in ELISA method,which was 88.51%.The sensitivity of G6 and G3SAT is 87.36%and 89.66%respectively.The sensitivity of G2 c-ELISA is 75.86%,and the sensitivity of G2 i-ELISA is the lowest,60.92%.(5)For S2 immunized sheep positive serum,G6 RBPT was the most sensitive,is 100%.G3 RBPT,G3 SAT,G5 c-ELISA,G4 b-ELISA have the same sensitivity,is 91.67%,and G2 i-ELISA was the least sensitive,for 39.58%.In this paper,the sensitivity and specificity of the kits produced by different manufacturers were compared by detecting different serums.Among them,G6 RBPT,G4 b-ELISA and G5 c-ELISA had the best sensitivity and specificity.
