| In recent years,with the improvement of living standards,the market demand for fragrant rice also showed an upward trend.However,the yield of aromatic rice varieties which used in current production is generally low,so it is more and more urgent to select high-quality and high-yield fragrant rice varieties.In this study,chewing method,KOH immersion method and aroma metabolite target analysis were used to identify the aroma traits of 161 strains of perennial rice PR107.,and the functional molecular markers of the known aroma genes were also used to detect the aroma traits.The full-length sequence analysis of the fragrance gene Badh2 was carried out.The main results are as follows:1.After simple sensory analysis of PR107 strain by chewing method and KOH immersion method,the PR107 strain of perennial rice could be identified as fragrant rice line,the difference rate between the two methods was 20.5%.2.The difference between chewing method and metabolite target analysis was also found by metabolite target analysis,and the difference rate was 50%.However,the results of three phenotypic analyses were in accordance with normal distribution.Among the three methods,some of the results can not be verified with each other.It may be more persuasive to use these three methods to verify rice flavor simultaneously in production practice.3.After using functional molecular markers to detect the PR 107 population,we found that the fragrance mutation site of PR107 strain was consistent with the common mutation site reported in the literature.The PR107 strain was only detected by electrophoresis with functional molecular markers for the mutation type of Badh2 gene,exon 7 8bp deletion in exon 7 and three SNP mutations"on chromosome 8 of rice,the results of polyacrylamide electrophoresis were different from the three control groups.Except for the heterozygosity of the two strains,PR107-65 and PR107-86,the others showed mutants(aromatic type)at this gene locus.4.The sequence analysis of Badh2 gene on chromosome 8 of PR 107 strain was compared with Oryza longistaminata showed that in PR107 strain,there was a 8bp deletion at exon 7,20382871,and the codon corresponding to proline(Pro)at position 253 changed from CCT to Terminator TAA,which may result in the accLumulation of aroma substances.lt is highly consistent with the results of previous studies.5.2-acetyl-1-pyrrolidine was also accumulated in rice grains which the results of gene sequencing is wild-type(non-aromatic)and control group,the concentration of 2-acetyl-1-pyrroline in non-aromatic rice was significantly lower than that in homozygous aromatic rice at 0.05 level(P = 0.0104).It was not ruled out that there were minor genes at other loci,which led to the accumulation of 2AP in PR107 Badh2 hybrid lines of non-aromatic rice.The results have certain reference value and theoretical basis for the practical application of fragrant rice breeding. |
